Efeito do extrato das folhas de manga Ubá (Mangifera indica L) e da mangiferina na modulação da expressão dos receptores CB1, PPARγ e adipocinas em ratos alimentados com dieta de cafeteria

Resumo: Obesity is a multifactorial disease strongly associated with adipogenesis, inflammation and metabolic syndrome. The distribution of body fat is an important factor, and specifically adipose tissue, visceral and subcutaneous seem to be the link between weight gain, adipogenesis, inflammation and metabolic risk. Adipocytes are involved in the regulation of energy balance and metabolism can be modulated by hormones, cytokines and nutrients, including adiponectin, leptin and resistin. And PPAR gamma CB1 receptors are involved in regulating the production of adipocytes, and insulin sensitivity in the inflammatory process. Activation of PPAR gamma triggers anti- inflammatory activities in several cell types by modulating the expression of pro- inflammatory genes. As for the CB1 receptors are directly related to obesity and other metabolic disorders, and, in certain cases behave as other pro-inflammatory and as anti- inflammatory agents. The mango leaf extract and mangiferin (main bioactive compound isolated from the extract), possess pharmacological antioxidant, anti-inflammatory, antiadipogênica, among others. The aim of the study was to evaluate the effect of leaf extract of mango and mangiferin isolated the same, prophylaxis and treatment in adipogenesis, through weight gain, biochemical and gene expression of CB1 receptors, PPAR gamma and the adipokines adiponectin, resistin and leptin in adipose tissue of rats fed a cafeteria diet. The mango leaf extract and mangiferin were administered to the animals by gavage at a dose of 40mg/kg and 250mg/kg respectively. Before the start of the experiments the animals were adapted for 8 days. The study was divided into two stages: prevention and treatment. Prophylaxis animals receberm the cafeteria diet, the leaf extract and mangiferin Mango together for eight days. In the treatment period, the animals received only cafeteria diet for 8 days to stimulate adipogenesis and then treated with the leaf extract of the sleeve, the mangiferin and CB1 receptor antagonists and PPAR gamma for 5 days. For weight gain, weight gain and food consumption were monitored daily. After each trial, the animals were euthanized, where blood was withdrawn to obtain the serum for performing the biochemical analysis of triacylglycerols, glucose, total cholesterol and its fractions (performed by colorimetric xkits) and adipose tissue (visceral and subcutaneous ) removed for analysis of gene expression of adipokines receivers and proposals (obtained by RT-PCR). At the trial concerning the prophylaxis, a significant decrease in food intake in the group receiving the mango leaf extract and weight of adipose tissue was significantly lower in the groups treated with the extract and mangiferin, compared to the control group. In the group of animals receiving extracts a significant reduction in the expression of CB1 receptor and resistin, adiponectin as presented a statistically higher expression compared to the control. In the group of animals receiving mangiferin, there was a significant increase in the expression of CB1 and a statistically significant decrease in expression of adipokines leptin and adiponectin. In the trial for the treatment groups treated with leaf extract and mangiferin Mango, significantly decreased the accumulation of adipose tissue and triglyceride levels while HDL levels increased significantly compared to the control group. In the group treated with the extract and the expression of the leptin receptor CB1 was statistically significantly higher, while adiponectin were significantly lower. The data showed a possible anti-inflammatory effect of mangiferin and antiadipogênico, since the expression of CB1 and PPAR gamma and adipokines resistin and leptin were significantly higher and that the expression of adiponectin decreased significantly compared to the control group. In this sense, the study showed the beneficial effect of the leaf extract of mango and mangiferin in the prophylaxis and treatment of adipogenesis, inflammation and metabolic risk factors associated with obesity and metabolic syndrome.

A distribuição da gordura corporal é um fator relevante, e especificamente os tecidos adiposos, visceral e subcutâneo, parecem ser o elo entre o aumento de peso, a adipogênese, a inflamação e os riscos metabólicos. Os adipócitos estão envolvidos na regulação do balanço energético e seu metabolismo pode ser modulado por hormônios, citocinas e nutrientes, entre eles a adiponectina, a leptina e a resistina. Os receptores CB1 e PPARγ estão envolvidos na regulação da produção de adipócitos, na sensibilidade à insulina e no processo inflamatório. A ativação do PPARγ desencadeia atividades anti-inflamatórias em vários tipos de células pela modulação da expressão de genes próinflamatórios. Já os receptores CB1 estão diretamente relacionados à obesidade e outras desordens metabólicas, sendo que, em determinadas situações comportam-se como próinflamatórios e em outras como anti-inflamatórios. O extrato da folha de manga e a mangiferina (principal composto bioativo isolado do extrato), possuem propriedades farmacológicas antioxidante, anti-inflamatória, antiadipogênica, dentre outras. O objetivo do estudo foi avaliar o efeito do extrato da folha de manga e da mangiferina isolada o mesmo, a profilaxia e o tratamento na adipogênese, por meio da evolução ponderal, parâmetros bioquímicos e expressão gênica dos receptores CB1, PPARγ e das adipocinas adiponectina, resistina e leptina no tecido adiposo de ratos alimentados com dieta de cafeteria. O extrato da folha de manga e a mangiferina foram administrados nos animais, por gavagem, na dose de 250mg/kg e 40mg/kg respectivamente. Antes do início dos experimentos os animais ficaram em adaptação por 8 dias. O estudo foi dividido em dois tempos: profilaxia e tratamento. Na profilaxia os animais receberm a dieta de cafeteria, o extrato da folha de manga e a mangiferina juntos por 8 dias. No período de tratamento, os animais receberam somente dieta de cafeteria por 8 dias para estimular a adipogênese e em seguida tratados com o extrato da folha de manga, a mangiferina e os antagonistas dos receptores CB1 e PPARγ por 5 dias. Para a evolução ponderal, o ganho de peso e o consumo alimentar viiiforam acompanhados diariamente. Após cada período experimental, os animais foram submetidos a eutanásia, onde o sangue foi retirado para obtenção do soro para a realização das análises bioquímicas de triacilgliceróis, glicose, colesterol total e suas frações (realizadas por kits colorimétricos) e o tecido adiposo (visceral e subcutâneo) removido para as análises da expressão gênica dos receptores e adipocinas propostas (realizados por RT-PCR). No período experimental referente à profilaxia, houve diminuição significativa no consumo alimentar no grupo que recebeu o extrato da folha de manga e o peso do tecido adiposo foi estatisticamente inferior nos grupos que foram tratados com o extrato e a mangiferina, comparados ao grupo controle. No grupo de animais que receberam extrato houve uma redução significativa na expressão da resistina e do receptor CB1, enquanto a adiponectina apresentou uma expressão estatisticamente superior comparada ao controle. No grupo de animais que receberam a mangiferina, houve um aumento significativo na expressão do CB1 e uma diminuição estatisticamente significativa na expressão das adipocinas adiponectina e leptina. No período experimental referente ao tratamento, os grupos tratados com o extrato da folha de manga e a mangiferina, diminuiram significativamente o acúmulo do tecido adiposo e os níveis de triacilgliceróis enquanto os níveis de HDL aumentaram significativamente comparados ao grupo controle. No grupo tratado com o extrato a expressão da leptina e do receptor CB1 foi estatisticamente superior, enquanto a adiponectina foi significativamente inferior. Os dados demonstraram um possível efeito anti-inflamatório e antiadipogênico da mangiferina, uma vez que a expressão dos receptores CB1 e PPARγ e as adipocinas leptina e resistina foram estatisticamente superiores e que a expressão da adiponectina diminuiu significativamente comparados ao grupo controle.
