Características bioquímicas de qualidade em soja: interação genótipo x ambiente, estimativa de correlação e medida de similaridade entre ambientes

Resumo: The multipurpose feature of soybean grain has generated a growing demand for cultivars with specific traits. To be able to meet market demands efficiently is important to characterize soybean cultivars in relation to chemical components concerning quality. The objectives of this work were to evaluate genotype x environment interaction of the soybean traits: lipoxygenases 1 (LOX 1) and 3 (LOX 3) specific activities, total protein, KOH-soluble protein, storage proteins ß-conglycinin subunits (α , α and ß) and glycinin subunits (acidic and basic) concentrations. In addition, the correlations between these traits in different cultivars and the degree of similarity among environments were estimated. Twenty-five soybean cultivars were evaluated at the COOPADAP experimental stations located in the districts of Bom Jesus de Goiás-GO, Paracatu-MG, São Gotardo-MG and Rio Paranaíba-MG, in the agricultural year 2005/2006. The experiments were set in randomized blocks, with three repetitions. Glycinin and ß-conglycinin storage proteins subunits contents were determined by SDSPAGE/ densitometry, total protein content by the Kjeldahl method, and soluble protein in 0.2% KOH. The coefficients of phenotypic correlation between the evaluated traits and the similarity degree among environments were estimated using Pearson s correlation. Cultivar x environment interactions were found for total protein, KOH-soluble protein and α subunit (ß- conglycinin) concentrations. Effects of cultivar and environment were found for LOX 1 and 3 specific activities, ß subunit (ß-conglycinin), acidic and basic subunits (glycinin) concentrations. Comparison among means to verify the effect of each cultivar in the different environments showed that genotypes Cs02020, Cs02054 and Cs02529 had superior performance for LOX 1 and 3 specific activities, ß subunit (ß- conglycinin) and basic subunits (glycinin) concentrations, whereas the means of α subunit (ß-conglycinin) and acidic subunits (glycinin) contents showed no significant difference among genotypes. The comparison among means to verify the effect of each environment upon genotypes showed that São Gotardo-MG had the largest means for LOX 1 and 3 specific activities, acidic, basic, α and ß subunits concentrations. Genotype x environment interaction studies showed that Cs02884, Luziania, Msoy8001, Cs02529 and Vencedora cultivars had superior performance for total protein, KOH- soluble protein and α subunit content in all tested environments. There was correlation between LOX 1 and 3 specific activities. The environments with higher similarity degree for LOX 1 specific activity were Paracatu-MG and Rio Paranaíba-MG, for LOX 3, Bom Jesus de Goiás-GO and São Gotardo-MG; for total protein and KOH-soluble protein Paracatu-MG and São Gotardo-MG; for ß subunit content Bom Jesus de Goiás-GO and Paracatu-MG. No correlations between environments were found for α , α and basic subunits contents. The genotypes appearing more frequently among the largest means for pairs of traits were Cs02936 and Luziania for LOX 1 and 3 specific activities and Cs9877 for KOH-soluble protein and α , α, ß, acidic and basic subunits contents.

Portanto, para melhor atender às exigências do mercado, torna-se importante a caracterização de cultivares de soja quanto aos componentes químicos relativos à qualidade. Os objetivos deste trabalho foram avaliar a interação genótipo x ambiente das características de qualidade do grão de soja: atividade específica de lipoxigenases 1 (LOX 1) e 3 (LOX 3), concentração de proteína, concentração de proteína solúvel em KOH, concentrações das subunidades da proteína de reserva β-conglicinina (α , α e β) e concentrações das subunidades da proteína de reserva glicinina (acídicas e básicas), além disso foram estimadas as correlações entre as características nos materiais genéticos e o grau de similaridade entre os ambientes de cultivo. Para isto, foram avaliados 25 genótipos de soja, cultivados nas estações experimentais da COOPADAP situadas nos municípios de Bom Jesus de Goiás-GO, Paracatu-MG, São Gotardo-MG e Rio Paranaíba-MG, no ano agrícola 2005/2006. Os experimentos foram executados em blocos casualizados, com três repetições. Foram determinados os conteúdos das subunidades das proteínas de reserva glicinina e β- conglicinina por SDS-PAGE/densitometria, conteúdo de proteína pelo método de Kjeldahl, atividade específica das lipoxigenases 1 e 3 e solubilidade protéica em KOH 0,2%. Os coeficientes de correlação fenotípicos entre os caracteres avaliados e o grau de similaridade entre ambientes foram estimados utilizando-se o método de correlação de Pearson. Verificou-se interação genótipo x ambiente para concentração de proteína, concentração de proteína solúvel em KOH e concentração da subunidade α (β-conglicinina). Verificou-se efeitos de genótipo e de ambiente para atividade específica de LOX 1 e 3, concentração da subunidade β (β-conglicinina) e subunidades acídicas e básicas (glicinina). Os estudos de comparação entre médias para verificar o efeito de cada genótipo no conjunto de ambientes permitiram concluir que os genótipos Cs02020, Cs02054 e Cs02529 se destacaram em relação aos demais apresentando um desempenho superior para atividade específica de LOX 1 e 3, concentração da subunidade β (β-conglicinina) e das subunidades básicas (glicinina) e que para as concentrações da subunidade α (β- conglicinina) e das subunidades acídicas (glicinina) não foram observadas médias estatisticamente diferentes entre si. Os estudos de comparação entre médias para verificar o efeito de cada ambiente no conjunto de genótipos apontaram o município de São Gotardo-MG como o que apresentou as maiores médias para as atividades específicas de LOX 1 e 3 e concentração das subunidades acídicas, básicas, α e β. Os estudos da interação genótipo x ambiente permitiram concluir que os genótipos Cs02884, Luziania, Msoy8001, Cs02529 e Vencedora se destacaram em relação aos demais apresentando desempenho superior para concentração de proteína, concentração de proteína solúvel em KOH e concentração da subunidade α em todos os ambientes avaliados. Observou-se correlação entre as atividades específicas de LOX 1 e 3. Verificou-se que os ambientes que apresentaram maior grau de similaridade para atividade específica de LOX 1 foram Paracatu-MG e Rio Paranaíba-MG e de LOX 3 foram Bom Jesus de Goiás-GO e São Gotardo-MG; para concentração de proteína e para concentração de proteína solúvel em KOH foram Paracatu-MG e São Gotardo- MG; para concentração da subunidade β foram Bom Jesus de Goiás-GO e Paracatu-MG e não foram encontradas correlações entre os ambientes para as concentrações das subunidades α , α e básicas.
