Atividade antiviral de extratos vegetais e flavonoides contra o Bovine herpesvirus 1 (BoHV1)

Resumo: The infections turn are had still as challenge for the public health and agricultural section. Several studies have as focus the discovery of new antiviral derived of natural products, mainly plants. The herpesvirus are one of the most used groups on this kind of research, due to importance for the public health and economical impact. Therefore, the purpose of this work was to evaluate the antiviral effects of flavonoids and three plants extract on the in vitro replication of bovine herpesvirus 1 (BoHV1). The the maximum non-cytotoxic concentration (CMNT) of DMSO and compounds it was measured by a variation of the method colorimeter based on the reduction of the salt MTT. The results showed that CMNT was 20 μg/mL for quercetina, 1.5 μg/mL for μg/mL and 1.5% for DMSO. For hexanic, dichloromethane, and etanolic extracts the values were 40, 25, and 60 μg/mL, respectively. Compounds' antiviral activity was evaluated through the rehearsal of determination of the virucidal activity and rehearsal of time of addition of the drugs in different stages of the cycle replicativo viral (timing of addition assays). DMSO did not present any activity in none of the accomplished experimental tests. All of substances presented significant virucidal effect. The quercetin and the dichloromethane extract demonstrated effect in the rehearsal of addition of the drug in the stages of adsorption andpenetration viral. The hexanic extract showed reduction of the title viral just in the phase of penetration stage. The ethanolic extract interfered on all stages of the replication in vitro of BoHV1, showing to be the most effective compound. The results of this work suggest the importance of the evaluated compounds for the veterinary and human medicine since they appear to be promising for the development of new antiviral therapies.

Diversos estudos têm como foco a descoberta de novos antivirais derivados de produtos naturais, principalmente vegetais. Os herpesvírus são uma classe muito utilizada nestas pesquisas, devido à importância para a saúde pública e impacto econômico. Neste contexto, o presente estudo busca avaliar o efeito in vitro de dois flavonoides e três extratos vegetais, na replicação in vitro do bovine herpesvirus 1 (BoHV1). A concentração máxima não tóxica (CMNT) do DMSO e dos compostos foi mensurada por uma variação do método colorimétrico baseado na redução do sal MTT. As CMNT obtidas foram 20μg/mL para quercetina, 1,5μg/mL para o TMDMB2, 1,5% (v/v) para o DMSO e 40, 25 e 60 μg/mL para os extratos hexânico, diclorometano e etanólico respectivamente. A atividade antiviral dos compostos foi avaliada por meio do ensaio de determinação da atividade virucida e ensaio de tempo de adição das drogas em diferentes etapas do ciclo replicativo viral (ensaio timing of addition). O DMSO não apresentou atividade antiviral em nenhum dos ensaios realizados. Todas as substâncias avaliadas apresentaram significante efeito virucida. A quercetina e o extrato diclorometano demonstraram efeito no ensaio de adição da droga nas etapas de adsorção e penetração viral. O extrato hexânico, mostrou redução do título viral apenas na fase de pentraçao viral. O extrato etanólico interferiu em todas as etapas da replicação in vitro do BoHV1, se mostrando o composto mais eficaz.
