Resumo: This work aimed to evaluate the effect of natural compounds on the reduction of blood cholesterol and triglycerides in an experiment with hypercholesterolemic rabbits. Two natural compounds were analyzed: the colorant curcumin and the flavonoid hesperidin, isolated and in combination. The test-substances were supplied at different doses of 20 and 40 mg, orally, in capsules. Hiperlipidemia in the rabbits was attained by supplying the diets with 1% crystalline cholesterol and 0.55 colic acid. Total cholesterol concentration increased considerably in the animals fed diet containing cholesterol + colic acid, when compared with the group fed only ration. The curcumin treatments at doses of 20 and 40 mg provoked a reduction of 50 and 53%, respectively, in blood cholesterol in the group with hiperlipidemia induced by cholesterol and colic acid and of 36 and 1% in the HDL-c parameter; thus, the 40 mg dose was the most effective. The calcic atorvastatin-treated group had a reduction of 36% in the total cholesterol levels and increased HDL-c in 5%. Triacylglycerol total concentration was 53.47%, higher in the animals fed diet containing cholesterol + colic acid than in the group fed only ration. Hipertriglyceridemia produced by diet rich in cholesterol + colic acid was significantly reduced in the animals treated with curcumin at 20 and 40 mg, with reduction percentages of 54 and 15% at the triacylglycerol levels, respectively. The hesperidin-treated group had a cholesterol reduction of 12% at the dose of 40 mg, but at the dose of 20 mg there was an increase of 12% in total cholesterol. The HDL-cholesterol levels were reduced in 9% at the dose of 20 mg and had an increase of 8% at the dose of 40 mg. The group treated with calcic-atorvastin had a reduction of 59% at the triacylglycerol levels. Also, the hesperidin-treated group showed a reduction of 50 and 43% at the doses of 40 and 20 mg, respectively. When the combination curcumin + hesperidin (20 mg) was used, a reduction of 24% in total cholesterol was verified, while the combination curcumin + hesperidin at the dose of 40 mg reduced total cholesterol in 9%. Both combinations led to an HDLcholesterol reduction of 13 and 25%, at the doses of 20 and 40 mg, respectively. The combinations curcumin + hesperidin had a smaller, though significant, reduction (36 and 42%) at the doses of 20 and 40 mg, respectively, than in the isolated doses for triacylglycerols. At the dose of 40 mg, the combination hesperidin +curcumin was more effective than in the isolated dose of curcumin at 40 mg. Overall, the natural compounds curcumin and hesperidin, except for hesperidin at 20 mg, presented pharmacological action rather accentuated on the lipid metabolism of hiperlipidemic rabbits, without, however, causing acute toxicity effects, as evidenced by the normal levels of TGO, TGP, GAMA GT, alkaline phosphatase and iron.

Foram estudados dois compostos naturais: o corante natural curcumina e o flavonoide hesperidina, isolados e de forma associada. As substâncias-teste foram fornecidas em diferentes doses de 20 e 40 mg, por via oral, em cápsulas. A hiperlipidemia dos coelhos foi alcançada suplementando-se as dietas com 1% de colesterol cristalino e 0,5% de ácido cólico. A concentração total de colesterol aumentou consideravelmente nos animais alimentados com dieta contendo colesterol + ácido cólico, quando comparada com o grupo que recebeu apenas ração. Os tratamentos com curcumina nas doses de 20 e 40 mg provocaram redução de 50 e 53%, respectivamente, no colesterol sanguíneo em relação ao grupo com hiperlipidemia induzida por colesterol e ácido cólico, e de 36 e 1% no parâmetro HDL-c; portanto, a dose de 40 mg foi mais eficaz. O grupo tratado com atorvastatina cálcica teve redução de 36% nos níveis de colesterol total e aumentou o HDL-c em 5%. A concentração total de triacilgliceróis foi 53,47% maior nos animais alimentados com dieta contendo colesterol + ácido cólico, em relação ao grupo que recebeu apenas ração. A hipertrigliceridemia produzida pela dieta rica em colesterol + ácido cólico foi significativamente reduzida nos animais tratados com curcumina a 20 e 40 mg, que tiveram porcentagens de redução de 54 e 15% nos níveis de triacilglicerol, respectivamente. O grupo tratado com hesperidina teve uma redução no colesterol de 12% na dose de 40 mg, porém na dose de 20 mg houve aumento no colesterol total em 12%. Os níveis de HDL-colesterol foram reduzidos em 9% na dosagem de 20 mg e apresentaram um aumento de 8% na de 40 mg. O grupo tratado com atorvastatina cálcica teve redução de 59% nos níveis de triacilglicerol. Também o grupo tratado com hesperidina mostrou redução de 50 e 43% nas doses de 40 e 20 mg, respectivamente. Quando se usou a associação curcumina + hesperidina (20 mg), verificou-se redução de 24% no colesterol total. Já a curcumina + hesperidina na dose de 40 mg reduziu o colesterol total em 9%. Ambas as associações provocaram a redução do HDL-colesterol em 13 e 25%, nas doses de 20 e 40 mg, respectivamente. As associações curcumina + hesperidina tiveram redução menor do que nas doses isoladas para triacilgliceróis, porém significativa (36 e 42%) nas doses de 20 e 40 mg, respectivamente. Na dose de 40 mg, a associação entre a hesperidina e a curcumina foi mais eficaz do que na dose isolada de curcumina a 40 mg.
