Comparação de kits comerciais na dosagem de constituintes bioquímicos do sangue em eqüinos hígidos

Resumo: Commercial kits of Bioclin® Company, Katal®, In Vitro- Human® and Labtest® were used to measure the biochemical constituents and to verify their influence on the values of the biochemical parameters average on 30 mangalarga marchador horses, of both sex, adults and healthy. For every clinic biochemical parameter, it was used three different techniques, each one of them represented by one commercial kit. For each electrolyte it was performed dosages through two different methodologies, flame photometry and selective electrode ion. For the marking protein of acute phase, the C reactive protein was chosen, since it is the most used for human medicine and for its dosage, three different methodologies were chosen: nephelometry, immunoturbodimetry and agglutination on latex. The kits used for these dosages are the same used for the routine dosages on clinic biochemical laboratories, electrolytes and protein of acute phase. It was determined the glucose (GLI) values, total protein (PPT), albumin (ALB), C reactive protein (PCR), total bilirubin (BT), urea (UR), creatinine (CREAT) , alkaline phosphatase (AFL), creation fosfocinase (CK), Y-glutamil transferase (GGT), aspartato aminotransferase (AST), sodium (Na+), potassium (K+), Clorets (CI-), total magnesium (Mg+2) and total calcium (tCa).There was no significant difference on albumin, urea, creatinine, glucose, aspartato aminotransferase, total bilirubin, potassium and sodium, there not having, therefore, restrictions for the use on auxiliary diagnostic on veterinarian medicine of anyone of the analyzed kits. On total protein dosages, the Katal® kit presented mean value of 7.72 mg/dl, higher than the mean values of Bioclin® and In Vitro-Human®. The alkaline phosphatase as measured through the kit of In Vitro-Human®, presented significant difference having mean value of 370.41U/L higher than mean values of Bioclin® and Katal®. For the creatino fosfo cinase, the kit of Katal® presented significant difference having mean value of 355.10 U/L, higher than the mean values of Bioclin® and Labtest®. The glutamil gamma transpherase enzyme, quantified by the kit of Katal®, got values of 23.80U/L, presented significant difference, having mean value higher than the mean values of Bioclin® and In Vitro-Human®. The Clorets ion presented the mean value of 94.20mEq/L, for the Labtest® Kit, considerably lower than the Bioclin® and In Vitro-Human® values. The total Calcium presented difference among the three kits used. The higher of them 11.66mg/dL, the one obtained with the In Vitro-Human® kit, 10.95mg/dL the one obtained with the Labtest® kit. Therefore, the Katal® kits for total protein and gamma glutamil transferase, from Labtest®, for clorets from In Vitro-Human®, for alkaline phosphatase and C reactive protein in latex from Bioclin®, must be used with caution when used as auxiliary diagnoses on horses.

Para cada parâmetro de bioquímica clínica foram utilizados três diferentes técnicas, cada uma delas representada por um kit comercial. Para cada eletrólito foram realizadas as dosagens através de duas metodologias distintas fotometria de chama e Íon eletrodo seletivo. Para as proteínas marcadoras de fase aguda, a proteína c reativa foi escolhida, por ser a mais utilizada em medicina humana, e para sua dosagem três metodologias diferentes foram escolhidas nefelometria, imunoturbidimetria e aglutinação em látex. Os kits utilizados nestas dosagens são os mesmos utilizados nas dosagens de rotina nos laboratórios para bioquímica clínica, eletrólitos e proteínas de fase aguda. Foram determinados os valores de glicose (GLI), proteínas totais (PPT), albumina (ALB), proteína c reativa (PCR), bilirrubina total (BT), uréia (UR), creatinina (CREAT), fosfatase alcalina (AFL), creatino fosfo cinase (CK), Y-glutamil transferase (GGT), aspartato amino transferase (AST), sódio (Na+), potássio (K+), cloretos (Cl-) magnésio total (Mg+2) e cálcio total (tCa). Não houve diferença significativa para os valores de albumina, uréia, creatinina, glicose, aspartato aminotransferase, bilirrubina total, potássio e sódio, não existindo, portanto, restrições para utilização no auxilio diagnóstico em medicina veterinária de qualquer um dos kits avaliados. Nas dosagens de proteínas totais, o kit da Katal® apresentou valor de média de 7,72mg/dL superior aos valores de média de Bioclin® e de In Vitro-Human®. A fosfatase alcalina quando mensurada através do kit da In Vitro- Human®, apresentou diferença significativa, com valor de média de 370,41U/L, superior aos valores de média de Bioclin® e Katal®. Para a Creatino fosfocinase o kit da Katal®, apresentou diferença significativa com valor de média de 355,10U/L, maior que os valores de média de Bioclin® e Labtest®. A enzima gamma glutamil transferase quantificada pelo kit da Katal® obteve valor de 23,80U/L, apresentou diferença significativa, com valor de média superior aos valores de média de Bioclin® e In Vitro-Human®. O íon cloreto apresentou o valor de média de 94,20mEq/L, para o kit da Labtest®, significativamente menor do que os valores de Bioclin® e In Vitro-Human®. O cálcio total apresentou diferença entre os três kits utilizados sendo o maior deles 11,66mg/dL o obtido com o kit da In Vitro-Human®, de 10,95mg/dL da Bioclin® e de 9,88mg/dL obtido com o kit de Labtest®.
