Resumo: This study aimed to evaluate the effects of the flavonoid naringin, hesperidin and chrysin and extract of Helianthus annuus in experimental colon carcinogenesis in rats exposed to 1.2 dimethylhydrazine. In each experiment we used Wistar female weighing 27.1 g ± 130.8 received weekly subcutaneous injection of 1,2-dimethylhydrazine (DMH, 20 mg/kg) for 10 weeks. From week 11, treatment was started with the flavonoid naringin, chrysin, hesperidin and the hydroalcoholic extract of Helianthus annuus, administered orally three times a week every 48 hours for 14 weeks. The animals were divided into 7 groups with five animals in each group. Group 1: NaCl 0,9%, Group 2: DMH + NaCl 0,9%, Group 3: DMH + treatment (10 mg/Kg), Group 4: DMH + treatment (100 mg/Kg), Group 5: DMH + treatment (200 mg/Kg). The evaluation of the results was reaizada from analyzes to quantify the number of aberrant crypt foci (ACF), histopathology, immunohistochemistry and histomorphometry, evaluation of nucleolar organizer regions per nucleus of enterocytes and enteroendocrine cells AgNORs, serum biochemical analysis and microanalysis minerals. Overall, the induction and control groups treated with the lower dose significantly increased the number of dysplastic crypts with dilated FCA, of AgNORs/nucleus and reduction of antioxidant minerals such as copper, magnesium, selenium and zinc compared to the healthy group. In other dosages used were a decrease in colorectal morphological disorganization and a decrease in the number of precancerous lesions in rats exposed to the chemical carcinogen DMH. The early and middle segments of the colon were more sensitive to the inhibitory effect of these treatments on the intestinal epithelial dysplasia, which apparently have limited influence on pre-established tumors. It is believed that some of the results may be due to the effects of these treatments in reducing the proliferation and recovery levels of minerals antioxidants in colonic mucosa.

Em cada experimento, foram utilizados ratos Wistar fêmeas pesando 130,8 ± 27,1g que receberam injeção subcutânea semanal de 1,2-dimetilhidrazina (DMH, 20 mg/kg) durante 10 semanas. A partir da 11ª semana, foi iniciado o tratamento com os flavonóides naringina, crisina, hesperidina e com o extrato hidroalcoólico de Helianthus annuus, administrado por gavagem 3 vezes por semana a cada 48 horas, durante 14 semanas. Os animais foram divididos em 5 grupos com 7 animais em cada grupo. Grupo 1: NaCl 0,9%; Grupo 2: DMH + NaCl 0,9%; Grupo 3: DMH + tratamento (10 mg/Kg); Grupo 4: DMH + tratamento (100 mg/Kg); Grupo 5: DMH + tratamento (200 mg/Kg). A avaliação dos resultados foi reaizada a partir de análises de quantificação do número de focos de criptas aberrantes (FCA), histopatologia, histoquímica e histomorfometria, avaliação de regiões organizadora de nucléolo por núcleo de enterócitos AgNORs e de células enteroendócrinas, análise bioquímica sérica e microanálise de minerais. De forma geral, nos grupos controle da indução e tratado com a menor dose houve aumento significativo do número de FCA com criptas displásicas dilatadas, das AgNORs/núcleo e redução dos minerais antioxidantes como cobre, magnésio, selênio e zinco comparado ao grupo sadio. Nas outras dosagens utilizadas foi observada redução da desorganização morfológica colorretal, com a diminuição do número de lesões pré-neoplásicas em ratos expostos ao carcinógeno químico DMH. Os segmentos inicial e médio do cólon foram mais sensíveis ao efeito inibitório destes tratamentos sobre a displasia do epitélio intestinal, os quais aparentemente apresentam influência limitada sobre lesões tumorais previamente estabelecidas.
