Caracterização molecular de linhagens de milho tropical por marcadores microssatélites

Resumo: | Knowledge of genetic diversity (GD) and relationships among maize inbred lines may allow characterization of the germplasm sort the genotypes into distinct heterotic groups, and this information is important in the choice of parents to obtain hybrids with high heterosis and increased grain yield. Our objectives in this study were (1) investigate the genetic diversity among tropical maize inbred lines from the germplasm collection of Maize Program® UFV,(2)estimate the potential informativeness of the SSR loci studied in maize inbred lines and (3) investigate the presence of alleles present in groups of strains belonging to different companies for improvement. The genetic similarity between all pairs of strains (ii ') were calculated by the weighted index, and for calculating the dissimilarity measure (Dii') we used the arithmetic complement (Dii'= 1 - S ii'). In total, 471 SSR alleles were identified, with an average of 5.81 alleles per locus. The average polymorphism information content (PIC) was 0. 5733. The genetic dissimilarity between strains ranged from 0.19 to 0.67, with an average of 0.49. Cluster analysis by the Tocher and UPGMA method was efficient in grouping inbred lines, suggesting a total of eighteen groups. Most of the inbred lines were consistent with its origins, although with some discrepancies. The pattern of groupings of the inbred lines revealed that there is ample genetic base among the majotity of inbred lines. The plausible ample genetic base among the majotity of inbred lines. The plausible explanation is that these were derived from different heterotic groups. The variability detected in the groups formed by SSR loci may contribute to the efficient use of inbred lines for exploitation of heterosis and broadening the genetic base of the program. Os objetivos deste estudo foram (1) investigar o nível de diversidade genética existente entre as linhagens de milho tropical do Banco de Germoplasma do Programa Milho, (2) estimar o potencial de informatividade dos locos SSRs nas linhagens de milho estudadas e (3) averiguar a presença de alelos exclusivos presentes em grupos de linhagens pertencentes a diferentes empresas de melhoramento. A similaridade genética entre todos os pares de linhagens (ii ) foi calculada pelo índice ponderado, e para o cálculo da medida de dissimilaridade (Dii ) utilizou-se o complemento aritmético (Dii = 1 S ii ). No total, 471 alelos SSR foram identificados, com uma média de 5,81 alelos por loco. O conteúdo informativo de polimorfismo (PIC) foi de 0,5733. A dissimilaridade genética entre as linhagens variou 0,19-0,67, com uma média de 0,49. A análise de agrupamento pelo método de Tocher e UPGMA foi eficiente no agrupamento das linhagens, sugerindo um total de dezoito grupos. A maioria das linhagens esteve condizente com suas origens, embora com algumas discrepâncias. O padrão de agrupamento das linhagens revelou haver uma base genética ampla entre a maioria das linhagens. Uma explicação plausível é que estas foram originadas de grupos heteróticos diferentes.
