Resumo: For many decades, studies have been developed in order to improve the biochemical and hemgasometric parameters of erythrocytes stored in bags of blood transfusion. Although there are still indications for the use of total blood, nowadays the balls are centrifuged and the blood components are separated to obtain in the erythrocytes concentrations (EC) and plasma. The viability of EC preserved by refrigeration is dependent on several factors. Among them the anticoagulant and the preservative solution added to the EC. Studies have shown that the addition of disodium phosphate (DP) preserves the pH in the best way, keeping it at higher levels which acts directly on the maintenance of adenosine triphosphate (ATP), a component involved in the preservation of the plasma membrane and thereby maintaining the normal shape of the cell and 2,3-DPG (2,3-DPG), a substance responsible for the release of oxygen to the tissues by hemoglobin. Methodology: In this study it was observed the biochemical and blood gas EC stored in CPD/SAG-M bags and CPD/SAG-M's added from DP. The bags were evaluated for 60 days. Results and Discussion: It was observed that the mediums of plasma potassium showed lower growth in the group treated with DP compared to the untreated group, even showing inversely proportional correlation to the behavior of pH in which it was observed averages always higher in the group added with the DP. Thus the best performance of the bags increased with DP in the maintenance of plasma membrane can be noticed. This finding is confirmed by a lower percentage of hemolysis suffered by the EC retained the balls containing the DP throughout the entire study. The consistently higher pH levels for the balls added with the FD show a higher rate of 2,3-DPG recovery, which showed higher average in the group treated with the DP. However, in the first day of the storage, the averages of 2,3-DPG were at zero, reaching maximum values at the 10th day of storage, demonstrating the capacity of producing 2,3-DPG in the canine erythrocytes even during the conservation process. The red cells preserved with DP showed lower morphological changes compared to the untreated group, also demonstrating a significant correlation with the potassium and 2,3-DPG variables, allowing to assume the average values of these biochemical variables from the morphological evaluation of stored erythrocytes. The erythrocytes of both groups showed an increase of the average diameter in the first day of the study, also with higher averages than those found in in vivo cells. From then it began to show reduction of the average diameter. It was concluded that the addition of DP to the CPD/SAG-M bags enhanced the preservation of erythrocyte up to the maximum period of 60 days of storage.

Embora ainda existam indicações para o uso do sangue total, atualmente as bolas são centrifugadas e separando-se os hemoderivados para a obtenção principalmente do concentrado de eritrócitos (CE) e plasma. A viabilidade do CE conservado sob refrigeração está na dependência de diversos fatores dentre eles o anticoagulante e a solução conservadora adicionada ao CE. Estudos têm demonstrado que a adição do Fosfato Dissódico (FD) melhor conserva o pH, mantendo este em níveis mais elevados o que age diretamente na manutenção do Trifosfato de Adenosina (ATP), componente envolvido na preservação da membrana plasmática e consequente manutenção do formato normal da célula, e do 2,3-Difosfoglicerato (2,3-DPG), substância responsável pela liberação do oxigênio aos tecidos pela hemoglobina. Foi Metodologia: Neste estudo foram observadas as alterações bioquímicas e hemogasométricas do CE conservado em bolsas CPD/SAG-M e CPD/SAG-M acrescidas do FD. As bolsas foram avaliadas por 60 dias. Resultados e Discussão: observado que as médias do potássio plasmático apresentaram menor crescimento no grupo tratado com o FD em comparação ao não tratado, inclusive demonstrando correlação inversamente proporcional ao comportamento do pH para o qual se observou médias sempre superiores no grupo acrescido do FD. Desta forma é possível constatar o melhor desempenho das bolsas acrescidas do FD na manutenção da membrana plasmática. Este dado é confirmado pelo menor percentual de hemólise sofrido pelo CE conservado nas bolas contendo o FD ao longo de todo o estudo. Os níveis constantemente superiores do pH para as bolas acrescidas do FD denotam maior taxa de recuperação do 2,3-DPG, que demonstrou médias superiores no grupo tratado com o FD. Entretanto, no primeiro dia de conservação as médias do 2,3-DPG estavam em zero, alcançando valores máximos ao 10º dia de conservação, demonstrando a capacidade de produção do 2,3-DPG dos eritrócitos caninos mesmo durante o processo de conservação. Os eritrócitos conservados com o FD apresentaram menores alterações morfológicas em comparação ao grupo não tratado, inclusive demonstrando correlação significativa com as variáveis potássio e 2,3-DPG, permitindo presumir os valores médios destas variáveis bioquímicas a partir da avaliação morfológica dos eritrócitos armazenados. Os eritrócitos de ambos os grupos apresentaram aumento do diâmetro médio já no primeiro dia do estudo, inclusive com médias superiores às encontradas nas células in vivo. A partir de então passou a apresentar redução do diâmetro médio.
