Dentre as principais complicações associadas ao DM2 encontra-se a retinopatia que, atualmente, é considerada uma das principais causas de cegueira em indivíduos adultos. O processo inflamatório crônico e de baixo grau presente na obesidade, favorece o descontrole glicêmico que, a longo prazo, contribui para o desencadeamento da retinopatia. Em linhas gerais, os ácidos graxos ômega-3 são amplamente investigados na literatura devido ao seu potencial anti-inflamatório. O mecanismo de ação do ômega-3 ocorre por meio de seus receptores específicos, GPR120/GPR40, que são capazes de desarticular a cascata inflamatória, levando a diminuição da inflamação em tecidos periféricos e no sistema nervoso central. Contudo, a presença e a ação desses receptores no tecido retiniano ainda é pouco descrita e investigada. Objetivo: Avaliar a presença e o papel dos receptores para ácidos graxos ômega-3 no controle do processo inflamatório da retina de animais obesos e resistentes à insulina. Metodologia: Camundongos Swiss machos, com 4 semanas de idade foram distribuídos em 2 grupos: controle (CT) e obeso (HF). O grupo controle foi mantido em ração comercial e o obeso exposto a dieta rica em gordura saturada por 9 semanas. Após esse período foi realizado teste de tolerância à insulina (TTI) para analisar a homeostase glicêmica. Posteriormente, os animais expostos a dieta rica em gordura foram distribuídos randomicamente nos grupos: dieta rica em gordura (HF) e dieta rica em gordura substituída em 10% de gordura por óleo de semente de linhaça, fonte de ômega-3 (HF+w3), por nove semanas. Ao final do período experimental, os testes de tolerância à insulina e a glicose foram realizados (TTI e TTG, respectivamente) e a retina foi extraída para análises da expressão gênica e do conteúdo proteico dos receptores para w3 (GPR120 e GPR40), de citocinas inflamatórias (TNF-alfa, IL-10 e IL-1ß), TAK1 e do marcador de retinopatia diabética (VEGF). Para análise fisiológica da função visual, foi utilizado o teste de eletrorretinografia. Resultados: Os animais expostos a dieta rica em gordura saturada (HF) apresentaram aumento da massa corporal, resistência à insulina e intolerância à glicose quando comparados ao grupo CT. Por outro lado, o grupo HF+?3 apresentou redução na glicemia de jejum e maior tolerância à glicose em comparação com o grupo HF. Em relação aos dados moleculares, mostramos que a retina dos animais era capaz de expressar os genes para os receptores GPR120 e GPR40, além disso, os animais expostos ao óleo de semente de linhaça apresentaram maior conteúdo proteico de GPR120 em relação ao grupo HF. Em relação às citocinas pró-inflamatórias, o grupo HF apresentou aumento na expressão gênica de TNF-alfa e IL-1ß, quando comparado ao CT. E o grupo HF+w3, redução na expressão gênica das citocinas quando comparados ao grupo HF. A respeito do conteúdo proteico, os animais do grupo HF apresentaram aumento no conteúdo proteico de IL-1?, p-TAK1 e VEGF quando comparado ao grupo CT. Além disso, o grupo HF+?3 apresentou redução no conteúdo proteico de TNF-alfa, IL-1ß, p-TAK e VEGF em relação ao grupo HF. No que se refere ao receptor GPR40 e a citocina IL-10, as diferente dietas experimentais não foram capazes de modular seu conteúdo proteico. Em relação ao teste funcional retiniano, animais do grupo HF apresentaram menor potencial oscilatório quando comparado ao grupo CT. Entretanto, a dieta rica em óleo de semente de linhaça foi capaz de proteger contra o desenvolvimento da retinopatia. Conclusão: A dieta high-fat foi capaz de induzir inflamação e danos na retina neural em camundongos Swiss. Por outro lado, a substituição em 10% por óleo de semente de linhaça foi capaz de reduzir a intolerância à glicose, expressão gênica e conteúdo proteico de marcadores inflamatórios e do fator de crescimento (VEGF). Sendo assim, nossos achados indicam que a dieta contendo óleo de semente de linhaça pode retardar o desenvolvimento de retinopatia diabética em camundongos obesos alimentados com dieta high-fatAbstract: Introduction: The prevalence of obesity has reached alarming levels, contributing to the development of several comorbidities specially type 2 diabetes (T2D). Among the main complications associated with DM2 lies retinopathy, which is currently considered one of the main causes of blindness in adults. The chronic and low-grade inflammatory process present in obesity favors the loss of glycemic control that, in the long term, contributes to the onset of retinopathy. In general, omega-3 fatty acids are widely investigated in the literature because of their anti-inflammatory potential. The mechanism of action of omega-3 fatty acids occurs through their specific receptors, GPR120 / GPR40, which are able to disarticulate the inflammatory cascade leading to decreased inflammation in peripheral tissues and the central nervous system. However, the presence and action of these receptors on retinal tissue is still poorly described and investigated. Objective: Evaluate the presence and role of the omega-3 fatty acids receptors in the control of the retinal inflammatory process of obese and insulin resistant animals. Methods: Four-week-old male Swiss were randomly distributed into two groups: control (CT) and obese (HF). The control group was maintained in standard chow diet and the obese group was exposed to a high fat diet for 9 weeks. After this period, an insulin tolerance test (ITT) was performed to analyze glucose homeostasis. After this, animals fed a high-fat diet were randomly distributed into two groups: maintained for another 9 weeks on HF diet (HF) or fed a diet substituted in 10% of lard to flaxseed oil, a source of omega-3 (HF+w3) for eight weeks. At the end of the experimental period, animals were submitted to an insulin and glucose tolerance tests (ITT, and GTT, respectively) and the retinas were extracted to analyze gene and protein expression of omega-3 fatty acids receptors (GPR120/GPR40), inflammatory cytokines (TNF-alfa, IL-10 e IL-1ß), TAK1 and the classical markers of diabetic retinopathy (VEGF). For the physiological analysis of visual function, we used a full flash electroretinography test. Results: The animals exposed to the high-fat diet (HF) presented an increase in body mass, insulin resistance and glucose intolerance when compared to the CT group. On the other hand, the HF+w3 group showed a reduction in fasting glucose and an improve in glucose tolerance compared to the HF group. In relation to the molecular data, we showed that animals retina were able of expressing the genes for GPR120 and GPR40 receptors and the animals exposed to the flaxseed oil showed an increase in GPR120 protein content compared to HF group. In relation to the proinflammatory cytokines, the HF group showed an increase in the gene expression of TNF-alfa and IL-1ß when compared to CT. The HF+w3 group showed a reduction in the cytokine gene expression when compared to the HF group. Regarding the protein content, the animals of the HF group showed an increase in the protein content of IL-1ß, p-TAK1 and VEGF when compared to the CT group. In addition, the HF+w3 group showed a reduction in the protein content of TNF-alfa, IL-1ß, p-TAK and VEGF in relation to the HF group. In relation to GPR40 receptor and IL-10 cytokine, the different experimental diets were not able to modulate their protein content. Regarding the retinal functional test, the animals of the HF group showed a decrease in the lower oscillatory potential when compared to the CT group. However, the diet rich in flax seed oil was able to protect against the development of retinopathy. Conclusion: The high-fat diet was able to induce inflammation and damage in the neural retina of Swiss mice. On the other hand, the substitution of 10% of lard to flaxseed oil was able to reduce glucose intolerance, gene expression and protein content of inflammatory markers and growth factors (VEGF).
