VIABILITY OF ENDOPHYTIC FUNGI FROM AMAZONIAN PLANTS PRESERVED BY THE CASTELLANI METHOD

Resumo: | The objective of this work was to evaluate the viability of endophytic fungi from Amazonian plants preserved by the technique of conservation in distilled water (Castellani) after two years of conservation. Endophytic fungi isolated from leaf and stem of Calycophyllum spruceanum, Carapa guianensis, Croton lechleri, Himatanthus sucuuba, Senna reticulata and Uncaria tomentosa stored by the Castellani technique for two years, were reactivated by the inoculation of three fungal fragments in PDA culture medium, incubated at 28oC for 14 days. After growth, the fungi were transferred to tubes for analysis of macromorphological characteristics. In total, 1050 endophytic fungi were reactivated. However, only 566 (53.9%) fungi were viable, 46.1% of endophytic fungi remained unviable due to contamination by fungi and bacteria (39.1%) and also due to non-growth of the samples (7%). Among the reactivated genera Trichoderma, Geotrichum, Cephaliophora and Rhizopus, showed viability of 100% of the samples. Reactivation rates for Acremonium, Aspergillus, Cladosporium, Fusarium, Penicillium, Phomopsis and Xylaria were greater than 50%. The Castellani method is effective for the preservation of various genera of fungal endophytes from plants in the Amazon. |

Fungos endofíticos isolados de folhas e caules de Calycophyllum spruceanum, Carapa guianensis, Croton lechleri, Himatanthus sucuuba, Senna reticulata e Uncaria tomentosa armazenados pela técnica de Castellani por dois anos, foram reativados pela inoculação de três fragmentos de fungos em meio de cultura BDA, incubado em 28oC por 14 dias. Após o crescimento, os fungos foram transferidos para tubos para análise das características macromorfológicas. No total, 1.050 fungos endofíticos foram reativados. Um total de 566 (53,9%) fungos eram viáveis, 46,1% dos endofíticos permaneceram inviáveis devido à contaminação por fungos e bactérias (39,1%) e também pelo não crescimento das amostras (7%). Dentre os gêneros reativados, Trichoderma, Geotrichum, Cephaliophora e Rhizopus, apresentaram viabilidade de 100% das amostras. As taxas de reativação para Acremonium, Aspergillus, Cladosporium, Fusarium, Penicillium, Phomopsis e Xylaria foram superiores a 50%. O método de Castellani é eficaz para a preservação de diversos gêneros de fungos endófitos de plantas na Amazônia.
