Evaluation of sexed semen deposition site on the in vivo embryo production of Nelore breed

Resumo: This study’s aim was to verify if the site of sexed semen deposition influences the quantity and quality of embryos from superovulated Nelore cows. Twelve females of proved ciclicity were used and went through SOV, FTAI and embryo collection protocols. FTAI was performed at 18 and 30h after the gonadorelin shot, with two straws each one containing 2,1x106 sperm cells, and the embryo collection was done seven days after the gonadorelin shot. Two repetitions were done, in a cross over design, according to semen deposition site, body or uterine horns. At the end of the treatment, 12.00 ± 2.95 and 12.25 ± 4.37 follicles were identified, with mean diameter of 8.64 ± 2.46 and 8.73 ± 2.42mm, for the groups inseminated in the uterine body and horns, respectively. It was noted that 66.7% (8/12) of the animals had CLs at the day of FTAI, presenting four animals in each group. The group inseminated in the uterine body had embryo recovery rate of 41.07% (23/56) and the group inseminated in the uterine horns had 35.71% (15/42). It was seen a decrease in the embryo recovery rate between repetitions, where the first repetition achieved 44.26% (27/61) while the second showed embryo recovery rate of 29.72% (11/37). Yielded 52.17% (12/23) of viable embryos for the group inseminated in the uterine body and 60% (9/15) for the group inseminated in the uterine horns. It was concluded that there weren’t differences at quantity or quality of recovered embryos according to the sexed semen deposition site at the time of FTAI.

Se utilizaron 12 hembras cíclicas, sometidas a los protocolos SOV y FTAI para la colecta de embriones. El IATF se realizó a las 18 y 30h de la administración de Gonadorelina, con dos pajuelas con una concentración de 2,1 x 106 de espermatozoides cada una, y los embriones se recolectaron siete días después de la aplicación de la gonadorelina. Se realizaron dos repeticiones, en diseño cruzado, según el lugar de deposición del semen, cuerpo o cuerno uterino. Al final del tratamiento superestimulador se cuantificaron 12,00 ± 2,95 y 12,25 ± 4,37 folículos, con un diámetro medio de 8,64 ± 2,46 y 8,73 ± 2,42 mm, en los grupos cuerpo y cuerno, respectivamente. Se observó que el 66,7% (8/12) de los animales tenían CL el día de la IATF, cuatro pertenecientes al grupo cuerpo y cuatro al grupo cuerno. El grupo de cuerpo mostró una tasa de recuperación embrionaria de 41,07% (23/56) y el grupo de cuerno, 35,71% (15/42). Se observó una disminución en la tasa de recuperación entre repeticiones, obteniendo 44,26% (27/61) en la primera repetición, mientras que en la segunda, 29,72% (11/37). Se recuperó el 52,17% (12/23) de embriones viables para el grupo de cuerpo y el 60% (9/15) para el grupo de cuerno. Se concluyó que no hubo disparidad en la cantidad o calidad de los embriones recuperados con respecto al lugar de depósito del semen sexado al momento de la IATF.Objetivou-se com este estudo descrever se o local de deposição do sêmen sexado influencia na quantidade e qualidade embrionária de vacas Nelore superovuladas. Foram utilizadas 12 fêmeas cíclicas, submetidas à protocolos de SOV e IATF para coleta de embriões. A IATF foi realizada 18 e 30h após a administração da Gonadorelina, com duas palhetas com concentração de 2,1 x 106 de espermatozoides cada, sendo a coleta dos embriões realizada sete dias após a aplicação da gonadorelina. Foram feitas duas repetições, em delineamento cross over, de acordo com o local de deposição do sêmen, corpo ou corno uterino. No final do tratamento superestimulatório foram quantificados 12,00 ± 2,95 e 12,25 ± 4,37 folículos, de diâmetro médio de 8,64 ± 2,46 e 8,73 ± 2,42 mm, nos grupos corpo e corno, respectivamente. Observou-se que 66,7% (8/12) dos animais apresentavam CLs no dia da IATF, sendo quatro pertencentes ao grupo corpo e quatro ao grupo corno. O grupo corpo apresentou taxa de recuperação embrionária de 41,07% (23/56) e o grupo corno, 35,71% (15/42). Foi observada diminuição da taxa de recuperação entre repetições, obtendo-se 44,26% (27/61) na primeira repetição enquanto na segunda, 29,72% (11/37).
