Autophagy and Cancer: a literature review

Resumo: Autophagia, or macroautophagy, is a phenomenon of cellular degradation and recycling that occurs constitutively at low levels in eukaryotic cells. Thasgained prominence as a pathway of mobilization of metabolic substrates that allow cancer cells to survive microenvironments with low nutrient supply or under conditions of oxidative stress, where the role of proteins such as beclin-1 and LC3B has been widely researched in various types of cancer. The aim of the present study was to review the literature on the performance of the autophagy process and its relationship with the development of cancer through autophagia. To design This article, a bibliographic survey was carried out covering the period from 1998 to 2019. Google academic, Scielo, LIlacs and Medline research portals were used. In this review, it is the result that autophagy has a dual role in cancer, as it can prevent tumor initiation through the suppression of chronic damage, inflammation, accumulation of damaged organelles and genomic instability, but Autophagia also maintains the Mitochondrial metabolic functions that provide nutrients for the growth of tumor cells, creating aggressive cancers.

Ha ganado prominencia como una vía de movilización de sustratos metabólicos que permiten a las células cancerosas sobrevivir a microambientes con bajo suministro de nutrientes o en condiciones de estrés oxidativo, donde el papel de proteínas como Beclin-1 y LC3B ha sido ampliamente investigado en varios tipos de cáncer.  Objetivo: El objetivo de este estudio fue revisar la literatura sobre el rendimiento del proceso de autofagia y su relación con el desarrollo del cáncer a través de la autofagia tumoral. Metodología: Para la concepción de este artículo, se llevó a cabo una encuesta bibliográfica que abarca el período comprendido entre 1998 y 2019. Se utilizaron los portales de búsqueda Google Scholar, Scielo, LIlacs y Medline.  Resultados y conclusión: Esta revisión resulta en la autofagia como un doble papel en el cáncer porque puede prevenir la iniciación del tumor a través de la supresión del daño crónico, inflamación, acumulación de orgánulos dañados e inestabilidad genómica, pero la autofagia también mantiene funciones metabólicas mitocondriales que proporcionan nutrientes para el crecimiento de las células tumorales, creando cánceres agresivos.Introdução: A autofagia, ou macroautofagia, é um fenômeno de degradação e reciclagem celular que ocorre constitutivamente em baixos níveis nas células eucarióticas. Tem ganhado destaque como via de mobilização de substratos metabólicos que permitem células cancerosas sobreviverem a microambientes com baixa oferta de nutrientes ou em condições de estresse oxidativo, onde o papel de proteínas como o Beclin-1 e LC3B tem sido amplamente pesquisado em diversos tipos de câncer. Objetivo: O objetivo do presente estudo foi fazer uma revisão de literatura acerca da atuação do processo de autofagia e sua relação com o desenvolvimento de câncer através da autofagia tumoral. Metodologia: Para concepção deste artigo foi realizado um levantamento bibliográfico que abrangeu o período de 1998 a 2019. Foram utilizados os portais de pesquisa Google Acadêmico, Scielo, LIlacs e Medline.
