Proliferative analysis of the Crandell-Rees Rim Feland cell (CRFK) grown with Dulbecco culture medium (DMEM) supplemented with B complex coenzymes

Resumo: The cell culture technique is used by all areas of science, mainly in Biotechnology, as it brings precise results and knowledge on new pathologies, studies on mechanisms of infections by microorganisms. The aim of this study was to analyze the proliferative potential of the Crandell-Rees Feline Kidney (CRFK) cell in static culture with Dulbecco culture medium (DMEM) supplemented with complex B coenzymes. This is a quantitative experimental study, divided into three groups: Control, Diverse and Diverse with B12, where the Control group did not receive extra concentration of coenzymes, the Diversas group received treatment with complex B coenzymes, which are: thiamine, riboflavin, niacin, pantathenic acid, pyridoxine, biotin and acid folic acid in four different concentrations, always doubling the concentration in the package insert of the DMEM culture medium, the third group received all seven coenzymes of the B complex plus cyanocobalamin (B12) that is not present in the package insert of DMEM. From this study it was concluded that the treatment with complex B coenzymes optimized the process of mitosis and subcultures time: in the Diverse group with B12 the culture time was 19.05 hours to reach the optimum point of confluence when compared Control group. Therefore, the study pointed out that coenzyme B12 is essential for cell metabolism, optimizing culture time by increasing the potential for cell mitosis.

El objetivo de este estudio fue analizar el potencial proliferativo de la célula del riñón felino de Crandell-Rees (CRFK) en cultivo estático con medio de cultivo Dulbecco (DMEM) suplementado con coenzimas complejas B. Este es un estudio experimental cuantitativo, dividido en tres grupos: Control, Diverse y Diverse con B12, donde el grupo Control no recibió concentración adicional de coenzimas, el grupo Diversas recibió tratamiento con coenzimas B complejas, que son: tiamina, riboflavina, niacina, ácido pantaténico, piridoxina, biotina y ácido ácido fólico en cuatro concentraciones diferentes, siempre duplicando la concentración en el inserto del paquete del medio de cultivo DMEM, el tercer grupo recibió las siete coenzimas del complejo B más cianocobalamina (B12) que no está presente en el inserto del paquete de DMEM. A partir de este estudio, se concluyó que el tratamiento con coenzimas B complejas optimizó el proceso de mitosis y tiempo de subcultivos: en el grupo Diverso con B12, el tiempo de cultivo fue de 19.05 horas para alcanzar el punto óptimo de confluencia en comparación Grupo de control. Por lo tanto, el estudio señaló que la coenzima B12 es esencial para el metabolismo celular, optimizando el tiempo de cultivo al aumentar el potencial de mitosis celular.A técnica de cultura celular é utilizada por todas as áreas das ciências, principalmente na área da Biotecnologia por trazer resultados precisos e conhecimentos sobre novas patologias, estudos sobre mecanismos de infecções por microrganismo. O objetivo deste estudo foi analisar o potencial proliferativo da célula Crandell-Rees Feline Kidney (CRFK) em cultura estática com meio de cultura Dulbecco (DMEM) suplementado com coenzimas do complexo B. Tratou-se de um estudo experimental quantitativo, dividido em três grupos: Controle, Diversas e Diversas com B12, onde o grupo Controle não recebeu concentração extra de coenzimas, o grupo Diversas recebeu tratamento com coenzimas do complexo B, sendo elas: tiamina, riboflavina, niacina, ácido pantatênico, piridoxina, biotina e ácido fólico em quatro concentrações distintas, sempre dobrando a concentração existente na bula do meio de cultura DMEM, o terceiro grupo recebeu todas as sete coenzimas do complexo B mais a cianocobalamina (B12) que não está presente na bula do DMEM. A partir deste estudo concluiu-se que, o tratamento com coenzimas do complexo B otimizou o processo de mitose e tempo das subculturas: no grupo Diversas com B12 o tempo de cultura foi de 19,05 horas para atingir o ponto ótimo da confluência quando comparada ao grupo Controle.
