New insights into the physicochemical properties of the human VMAT2 monoamine transporter and its mode of interaction with the neurotransmitter serotonin: An in silico analysis

Resumo: VMAT2 are glycoproteins capable of carrying monoamines from presynaptic vesicles to synaptic clefts during neuronal firing. Present in many species of animals including, mammals, reptiles and birds, this protein has been studied extensively, however, little is known about its physical-chemical characteristics and mode of interaction with native ligands or not. In order to better characterize human VMAT2, the present study was developed in order to explore various physical-chemical, biochemical and structural parameters related to this neurotransporter through in silico tools. In this work, new and relevant ideas about the structure and its mechanism of interaction with 5-HT are presented.

Presente en muchas especies de animales, incluidos mamíferos, reptiles y aves, esta proteína se ha estudiado ampliamente, sin embargo, se sabe poco sobre sus características fisicoquímicas y el modo de interacción con ligandos nativos o no. Para caracterizar mejor el VMAT2 humano, el presente estudio se desarrolló para explorar diversos parámetros físico-químicos, bioquímicos y estructurales relacionados con este neurotransportador a través de herramientas in silico. En este trabajo, se presentan ideas nuevas y relevantes sobre la estructura y su mecanismo de interacción con 5-HT.As VMAT2 são glicoproteínas capazes de carrear monoaminas a partir de vesículas pré-sinápticas para as fendas sinápticas durante disparos neuronais. Presentes em muitas espécies de animais incluindo, mamíferos, répteis e aves, esta proteína tem sido estudada extensivamente, porém, pouco se sabe sobre suas características físico-químicas e modo de interação com ligantes nativos ou não. Com o intuito de melhor caracterizar a VMAT2 humana, o presente estudo foi desenvolvido a fim de explorar vários parâmetros físico-químicos, bioquímicos e estruturais relacionados a esse neurotransportador por meio de ferramentas in silico.
