Avoidance test with earthworms (Eisenia andrei) in natural soil treated with a Bacillus thuringiensis-based biopesticide to soil quality evaluation

Resumo: Tests using earthworms is an important preliminary screening tool in soil ecotoxicology studies. The aim was to evaluate the natural soil quality containing Bacillus thuringiensis (Bt) as an agent in a biopesticide using Eisenia andrei as bioindicator. The filter contact test was based on OECD 207 guideline and avoidance test based on ISO 17512-1, with modifications. After exposure by contact, no mortality was observed, however, excess mucus released lethargic movements and decreased touch reflexes were observed from 100 mg L-1. These results raise the question of whether sublethal effects are relevant when about the physiological effects that would harm organism and their important functions in the ecosystem. In the avoidance test, there was no response to Bt soil, suggesting that the biopesticide does not alter the soil quality or present high toxicity to E. andrei. The use of Bt biopesticides indicates to few negative effects to annelids, what suggests, at least, a viable alternative in relation to the high toxicity of chemical insecticides frequently used in agriculture.

El objetivo del estudio fue evaluar la calidad del suelo que contiene Bacillus thuringiensis (Bt) como ingrediente activo de un bioplaguicida, utilizando Eisenia andrei como bioindicador. La prueba de contacto en papel de filtro se basó en el estándar OECD 207 y la prueba de fugas se basó en ISO 17512-1, con modificaciones. Después de un período de exposición por contacto, no hubo mortalidad, sin embargo, se observó un exceso de moco liberado, movimientos letárgicos y disminución de los reflejos al tacto a 100 mg L-1. Estos resultados plantean la cuestión de si los efectos subletales son relevantes al pensar en los efectos fisiológicos que dañarían al organismo y sus funciones importantes en el ecosistema. En la prueba de fuga, no hubo respuesta a Bt en el suelo, lo que sugiere que el bioplaguicida no altera la calidad del suelo o tiene una alta toxicidad para E. andrei. El uso de Bt en bioplaguicidas indica pocos efectos negativos para los anélidos, lo que sugiere, al menos, una alternativa viable en relación con la gran toxicidad de los insecticidas químicos utilizados con frecuencia en la agricultura.Teste de fuga utilizando minhocas é uma importante ferramenta em estudos ecotoxicológicos preliminares da qualidade do solo. O objetivo do estudo foi avaliar a qualidade do solo contendo Bacillus thuringiensis (Bt) como princípio ativo de um biopesticida, utilizando Eisenia andrei como bioindicador. O teste de toxicidade de contato em papel filtro foi baseado na norma OECD 207 e o teste de fuga foi baseado na ISO 17512-1, com modificações. Após período de exposição de contato não houve mortalidade, entretanto, excesso de muco liberado, movimentos letárgicos e diminuição dos reflexos aos toque foram observados em 100 mg L-1. Estes resultados levantam a questão para entender se os efeitos subletais são relevantes quando se pensa nos efeitos fisiológicos que prejudicariam o organismo e em suas importantes funções no ecossistema. No teste de fuga, não houve resposta ao Bt no solo, sugerindo que o biopesticida não altera a qualidade do solo ou apresenta toxicidade alta para E. andrei.
