Sickness behavior in obese mice

Resumo: The consumption of a hypercaloric diet leads to an increase in obesity, which can lead to an inflammatory state. Thus, the objective of this work was to analyze the influence of this inflammation on the behavior of obese animals. For this, male Swiss mice were kept under controlled feeding were divided into 2 groups; Group Cafeteria diet, which received high calorie foods and standard chow for rodent, while the control group received only standard rat chow. After 30 days, 2 hours before the behavioral tests, both groups were subdivided, so that half of the animals of each group received LPS by intraperitoneal injection (100 g/kg; i.p.) and the other half received saline injection ( 0.9% NaCl, 100g/kg; i.p.) (Groups: Cafeteria Diet + LPS and Cafeteria Diet + Saline; Normal Diet + LPS and Normal Diet + Saline). Afterwards, they were submitted to the Open Field Test and the Forced Swim Test and later euthanized for collection and weighing of the epididymal and retroperitoneal adipose tissue. There was an increase in weight, epididymal and retroperitoneal fat in mice treated with cafeteria diet compared to normal diet. LPS had a more pronounced effect in the cafeteria diet group, with a decrease in the number of entries in the periphery and in the center, in addition to the total number of entries and rearing, in the open field test. There was also an increase in time floating in the forced swim test. Therefore, the accumulation of body fat, by the ingestion of a hypercaloric diet, potentiates the inflammatory state caused by LPS, leading to an anxiogenic effect.

Por lo tanto, el objetivo de este trabajo era analizar la influencia de esta inflamación en el comportamiento de los animales obesos. Para ello, los animales se dividieron en 2 grupos; El grupo que recibió dieta de la cafetería y el grupo de control que recibió solo alimento comercial. Después de 30 días, ambos grupos se subdividieron, de modo que la mitad de los animales en cada grupo recibieron inyección intraperitoneal de LPS (100 g/kg; i.p.) y la otra mitad inyección de salina (0,9% de NaCl, 100 g/100 g/kg; i.p.) (Grupos: dieta de cafetería+LPS; dieta de cafetería+Salina; dieta normal+LPS; dieta normal+Salina). Posteriormente, fueron sometidos al Test de Campo Abierto y al Test de Nado Forzado y sacrificados para la colecta y pesaje del tejido adiposo epididimario y retroperitoneal. Hubo un aumento de peso, epidídimo y grasa retroperitoneal en los ratones tratados con dieta de cafetería. LPS tuvo un efecto más pronunciado en el grupo de dieta de cafetería, con una disminución en el número de entradas en la periferia, en el centro y del número total de entradas y rearings, en la prueba de campo abierto. También hubo un aumento en el tiempo de flotación en la prueba de nado forzado. Por tanto, la acumulación de grasa corporal, por la ingestión de una dieta hipercalórica, potencia el estado inflamatorio provocado por el LPS, dando lugar a un efecto ansiogénico.O consumo de dieta hipercalórica leva a um aumento de obesidade, podendo levar a um estado inflamatório. Assim, o objetivo desse trabalho foi analisar a influência dessa inflamação no comportamento de animais obesos. Para tanto, camundongos swiss machos adultos mantidos sob alimentação controlada, foram divididos em 2 grupos; o Grupo Dieta de Cafeteria, que recebeu alimentos de alto valor calórico e ração padrão e Grupo Controle tratado apenas ração comercial. Após 30 dias, 2 horas antes da realização dos testes, ambos os grupos foram subdivididos, de forma que, metade dos animais de cada grupo recebeu injeção intraperitoneal de LPS (100 g/kg; i.p.) e a outra metade injeção de salina (0.9% NaCl, 100g/Kg; i.p.) (GRUPOS: Dieta de Cafeteria+LPS e Dieta de Cafeteria+Salina; Dieta Normal+LPS e Dieta Normal+Salina). Os animais dos quatro grupos foram submetidos ao Teste do Campo Aberto e Teste do Nado Forçado e em seguida eutanasiados e os tecidos adiposo epididimal e retroperitoneal removidos para análise. Observou-se aumento de peso, gordura epididimal e retroperitoneal nos camundongos tratados com dieta de cafeteria. O LPS exerceu efeito mais acentuado no grupo de dieta de cafeteria, com diminuição do número de entradas na periferia e no centro, além do número total de entradas e rearings, no teste de campo aberto. Houve também aumento do tempo de imobilidade no teste de nado forçado.
