Immunohistochemical and histomorphometric analysis of the endometrium of rats in permanent estrus treated with melatonin, metformin and clomiphene citrate

Resumo: | Objective: To evaluate the effects of melatonin, metformin and clomiphene citrate on the endometrium of polycystic ovary syndrome model rats. Methods: Seventy-eight rats were divided into 13 groups and two experiments (prevention and treatment). After exposure, all animals were anesthetized and the uterine horns were removed and submitted to hematoxylin and eosin staining and immunohistochemistry (Ki-67 and Vegf). The data obtained were submitted to Student's t test (p<0.05). Results/Conclusion: Melatonin, metformin and clomiphene citrate do not promote proliferative and angiogenic effects on the endometrium of rats in permanent estrus, however, melatonin has a more significant trophic effect on the endometrial stroma, which did not interfere with its thickness, while that metformin had a more significant trophic effect on endometrial glandular epithelium, therefore, these effects are similar both before and after PCOS induction by continuous light exposure. |

Métodos: Setenta y ocho ratas se dividieron en 13 grupos y dos experimentos (prevención y tratamiento). Después de la exposición, todos los animales fueron anestesiados y los cuernos uterinos fueron removidos y sometidos a tinción con hematoxilina y eosina e inmunohistoquímica (Ki-67 y Vegf). Los datos obtenidos se sometieron a la prueba de la t de Student (p<0,05). Resultados/Conclusión: La melatonina, la metformina y el citrato de clomifeno no promueven efectos proliferativos y angiogénicos sobre el endometrio de ratas en celo permanente, sin embargo, la melatonina tiene un efecto trófico más significativo sobre el estroma endometrial, que no interfiere con su espesor, mientras que la metformina tuvo un efecto trófico más significativo sobre el epitelio glandular endometrial, por lo tanto, estos efectos son similares tanto antes como después de la inducción del SOP por exposición continua a la luz.Objetivo: Avaliar os efeitos da melatonina, metformina e citrato de clomifeno no endométrio de ratas modelo de síndrome dos ovários policísticos. Métodos: Setenta e oito ratas foram divididas em 13 grupos e dois experimentos (prevenção e tratamento). Após as exposições todos os animais foram anestesiados e tiveram retirados os cornos uterinos e submetidos a coloração pela hematoxilina e eosina e imunoistoquímica (Ki-67 e Vegf). Os dados obtidos foram submetidos ao teste t de Student (p<0,05).
