Polymorphisms in the IL17A gene do not influence the clinical, laboratory and radiological manifestations of tuberculosis

Resumo: | Some single-base polymorphisms (SNPs) in cytokine genes have the potential to affect gene expression and/or biological activity of the respective cytokine, and there is evidence of the association of different SNPs with tuberculosis in several different populations. Studies involving the influence of SNPs on the IL17A gene are still controversial, behaving differently among different populations. Thus, research into the association of SNPs in the IL17A gene with the clinical course of the disease may contribute to a better understanding of TB susceptibility and development. Therefore, the aim of the present study was to evaluate the influence of SNPs on the IL17A gene and their clinical-laboratory association in patients with tuberculosis. Therefore, 35 patients with tuberculosis and 26 controls were studied. The evaluation of the SNPs in the IL17A gene, rs7747909 and rs2275913, was performed using the allelic discrimination technique by real-time PCR. Clinical, laboratory and radiological data were obtained by collecting medical records. Analyzes were performed using Fisher's test, considering significant values p<0.05. There was no association of SNPs in the IL17A gene evaluated, either in relation to susceptibility or in terms of clinical, laboratory and radiological association. Therefore, it is essential to advance in research to analyze the influence of other SNPs on the IL17A gene and the relationship with other variables linked to the immune response to M. tuberculosis. |

Los estudios que involucran la influencia de los SNP en el gen IL17A aún son controvertidos y se comportan de manera diferente entre diferentes poblaciones. Por lo tanto, la investigación sobre la asociación de SNP en el gen IL17A con el curso clínico de la enfermedad puede contribuir a una mejor comprensión de la susceptibilidad y el desarrollo de la TB. Por tanto, el objetivo del presente estudio fue evaluar la influencia de los SNPs en el gen IL17A y su asociación clínico-laboratorio en pacientes con tuberculosis. Por tanto, se estudiaron 35 pacientes con tuberculosis y 26 controles. La evaluación de los SNPs en el gen IL17A, rs7747909 y rs2275913, se realizó mediante la técnica de discriminación alélica por PCR en tiempo real. Los datos clínicos, de laboratorio y radiológicos se obtuvieron mediante la recopilación de historias clínicas. Los análisis se realizaron mediante la prueba de Fisher, considerando valores significativos p<0,05. No hubo asociación de SNPs en el gen IL17A evaluado, ni en relación a la susceptibilidad ni en términos de asociación clínicos, de laboratorio y radiológicos. Por ello, es fundamental avanzar en la investigación para analizar la influencia de otros SNPs sobre el gen IL17A y la relación con otras variables ligadas a la respuesta inmune frente a M. tuberculosis.Alguns polimorfismos de base única (SNP) nos genes das citocinas apresentam potencialidade para afetar a expressão gênica e/ou a atividade biológica da respectiva citocina, e existem evidências da associação de diferentes SNPs com a tuberculose em diversas populações diferentes. Os estudos envolvendo a influência de SNPs no gene da IL17A ainda são controversos, comportando-se de forma variada entre diferentes populações. Desta forma, a pesquisa da associação de SNPs no gene da IL17A com a evolução clínica da doença, poderá contribuir para um melhor entendimento da susceptibilidade e o desenvolvimento da TB. Para tanto, o objetivo do presente estudo foi avaliar a influência de SNPs no gene IL17A e a sua associação clínico-laboratorial em pacientes com tuberculose. Para tanto, foram estudados 35 pacientes com tuberculose e 26 controles. A avaliação dos SNPs no gene IL17A, rs7747909 e rs2275913, foi realizada através da técnica de discriminação alélica por PCR em tempo real. Os dados clínicos, laboratoriais e radiológicos foram obtidos através da coleta de prontuário. As análises foram realizadas pelo teste de Fisher, considerando valores significativos p<0,05. Não houve associação dos SNPs no gene IL17A avaliados, seja em relação à susceptibilidade ou quanto à associção clínica, laboratorial e radiológica.
