Effects of Ximenia americana L. extract associated with corticoid and exercise in the treatment of chronic obstructive pulmonary disease induced in Rattus norvegicus

Resumo: | This research aims to observe the influence of aerobic exercise associated with the use of corticosteroids and aqueous extract of the stem bark of Ximenia americana L. in the treatment of Chronic Obstructive Pulmonary Disease. Three doses of papain via the nasal route were used in seven groups of male Wistar rats in order to induce pulmonary inflammatory response. The groups were divided into G1 (exercise + inflammation + steroids), G2 (exercise + inflammation), G3 (inflammation + exercise + X. americana), G4 (inflammation + X. americana), G5 (exercise + inflammation + X. americana + steroids), G6 (inflammation) and G7 (control). After the animals were euthanized, bronchial-alveolar lavage was performed, and the heart and left lung were collected for histomorphological analysis (H.E., P.A.S. and Masson's trichrome stains). It was found that the treatments performed in groups G1, G2, G3, G4, G5 were successful when compared to G7, with an equivalence of the treated groups in terms of RV wall thickness in relation to G6. Furthermore, through the preparations stained with H.E. from treated animals, dilated air spaces and small infiltration of inflammatory cells are evident. Qualitative analysis of slides stained by P.A.S. shows that there is a smaller number of mucus-producing goblet cells in preparations from rats exposed to a combination of treatment regimens, compared to groups in which there was only one form of treatment. The aqueous extract of X. americana stem bark had a significant contribution in the treatment of COPD, as well as aerobic exercise. |

Se utilizaron tres dosis de papaína por vía nasal en siete grupos de ratas Wistar macho para inducir una respuesta inflamatoria pulmonar. Los grupos se dividieron en G1 (ejercicio + inflamación + esteroides), G2 (ejercicio + inflamación), G3 (inflamación + ejercicio + X. americana), G4 (inflamación + X. americana), G5 (ejercicio + inflamación + X. americana) + esteroides), G6 (inflamación) y G7 (control). Después de la eutanasia de los animales, se realizó un lavado bronquial-alveolar y se recogieron el corazón y el pulmón izquierdo para análisis histomorfológico (tinciones H.E., P.A.S. y tricrómico de Masson). Se encontró que los tratamientos realizados en los grupos G1, G2, G3, G4, G5 fueron exitosos en comparación con G7, con una equivalencia de los grupos tratados en términos de espesor de la pared del VD en relación con G6. Además, a través de las preparaciones teñidas con H.E. de los animales tratados, se evidencian espacios de aire dilatados y una pequeña infiltración de células inflamatorias. Análisis cualitativo de portaobjetos teñidos por P.A.S. muestra que hay un número menor de células caliciformes productoras de moco en preparaciones de ratas expuestas a una combinación de regímenes de tratamiento, en comparación con los grupos en los que solo había una forma de tratamiento. El extracto acuoso de corteza de tallo de X. americana tuvo una contribución significativa en el tratamiento de la EPOC, así como en el ejercicio aeróbico.Essa pesquisa tem como objetivo observar a influência do exercício aeróbico associado ao uso de corticoide e extrato aquoso das cascas do caule de Ximenia americana L. no tratamento da Doença Pulmonar Obstrutiva Crônica. Utilizaram-se três doses de papaína via nasal em sete grupos de ratos Wistar machos a fim de induzir resposta inflamatória pulmonar. Os grupos foram divididos em G1 (exercício + inflamação + corticoide), G2 (exercício + inflamação), G3 (inflamação + exercício + X. americana), G4 (inflamação + X. americana), G5 (exercício + inflamação + X. americana + corticoide), G6 (inflamação) e G7 (controle). Após a eutanásia dos animais, realizou-se o lavado brônquico-alveolar, e coleta do coração e pulmão esquerdo para análises histomorfológicas (colorações em H.E., P.A.S e Tricromio de Masson). Verificou-se que houve o sucesso dos tratamentos realizados nos grupos G1, G2, G3, G4, G5 ao serem comparados com G7, com uma equivalência dos grupos tratados quanto à espessura da parede do VD em relação ao G6. Além disso, através das preparações coradas com H.E. provenientes de animais tratados, evidencia-se espaços aéreos dilatados e pequena infiltração de células inflamatórias. A análise qualitativa das lâminas coradas pelo P.A.S. mostra que há um número menor de células caliciformes produtoras de muco em preparações provenientes de ratos expostos a uma associação de esquemas de tratamentos, comparando-se com grupos em que houve uma única forma de tratamento.
