Phospholipases A2 from ofidic venoms: Update on purification, biological characterization and biotechnological application

Resumo: Ophidism is responsible for about 1.8 to 2.7 million cases of poisoning every year, and understanding the biological mechanisms responsible for its neurotoxicity is essential for therapeutic management. Among the diversity of molecules that compose snake venom there are the phospholipase A2, an enzyme family responsible for the production of a range of pathological conditions, including myotoxicity, cardiotoxicity, and neurotoxicity. The present work, carried out through the methodology of integrative literature review, aims to identify and indicate the main advances in the last 10 years regarding the characterization and understanding of the neurotoxic action of phospholipases A2 originating from snake venom, in the field of toxicology.  The main progress identified corresponded to the understanding that the neurotoxic activity of B. bilineata smargadine is based on the action of PLA2 Bbil-TX, proof of the dependence of PhTX-II by calcium for its action, identification of the occurrence of the muscular neurotoxic action of BP-13 in the sarcolemma region, determination of the subunit of crotoxin acting as a regulator of the GLIC receptor, and identification of the selectivity of MiDCA1 by specific potassium channels. The advance in the understanding of this enzymatic group corresponds to part of the pathway necessary for the development of therapeutics targeting PLA2 and possible biotechnological applications of these biomolecules.

Entre la diversidad de moléculas que componen el veneno de serpiente se encuentra la fosfolipasa A2, una familia de enzimas responsable de la producción de una serie de condiciones patológicas, como la miotoxicidad, la cardiotoxicidad y la neurotoxicidad. El presente trabajo, realizado a través de la metodología de revisión bibliográfica integradora, tiene como objetivo identificar e indicar los principales avances en los últimos 10 años respecto a la caracterización y comprensión de la acción neurotóxica de las fosfolipasas A2 del veneno de serpiente, en el ámbito de la toxicología.  Los principales avances identificados correspondieron a la comprensión de que la actividad neurotóxica de B. bilineata smargadine se basa en la acción de PLA2 Bbil-TX, la prueba de la dependencia de PhTX-II por el calcio para su acción, la identificación de la ocurrencia de la acción neurotóxica muscular de BP-13 en la región del sarcolema, la determinación de la subunidad de crotoxina que actúa como regulador del receptor GLIC y la identificación de la selectividad de MiDCA1 por canales de potasio específicos. El avance en el conocimiento de este grupo enzimático corresponde a parte de la ruta necesaria para el desarrollo de terapias dirigidas a PLA2 y a posibles aplicaciones biotecnológicas de estas biomoléculas.O ofidismo é responsável por cerca de 1,8 a 2,7 milhões de casos de envenenamentos ao ano, sendo a compreensão dos mecanismos biológicos responsáveis pela sua neurotoxicidade imprescindível para o manejo terapêutico. Entre a diversidade de moléculas que compõem a peçonha das serpentes existem as fosfolipase A2, família enzimática responsável pela produção de uma gama de quadros patológicos, incluindo a miotoxicidade, cardiotoxicidade e neurotoxicidade. O presente trabalho, realizado por meio da metodologia de revisão integrativa de literatura, visa identificar e indicar os principais avanços dos últimos 10 anos referentes à caracterização e o entendimento da ação neurotóxica originada das fosfolipases A2 provindas da peçonha de serpentes, no que tange o campo da toxicologia.  Os principais progressos identificados corresponderam a compreensão de que a atividade neurotóxica da B. bilineata smargadina é fundamentada na ação da PLA2 Bbil-TX, comprovação da dependência da PhTX-II por cálcio para sua ação, identificação da ocorrência da ação neurotóxica muscular da BP-13 na região do sarcolema, determinação da subunidade da crotoxina atuante como regulador do receptor GLIC e identificação da seletividade da MiDCA1 por canais específicos de potássio.
