Nanopartículas de prata associada ao trimetafosfato de sódio e fluoreto: síntese e avaliação do efeito antimicrobiano

Resumo: | Several researches have been conducted in order to develop new antimicrobial agents. Among these agents, silver nanoparticles (Ag-NP) demonstrated antimicrobial and antifungal activity against a broad spectrum of microorganisms. In addition, sodium trimetaphosphate (TMP) preserves the integrity and stability of the enamel and its association in appropriate proportions with fluoride (F) may provide a greater protective effect over conventional fluoride products. Thus, this study aimed to synthesize a compound containing silver nanoparticles (Ag-NP), sodium trimetaphosphate (TMP) and fluoride (F), characterize it and evaluate the antimicrobial action against planktonic cells of Candida albicans and Streptococcus mutans. For this, Ag-NP were synthesized by reducing silver nitrate at 1 (T1) or 10% (T10) in relation to TMP by a chemical route in the presence of F and TMP. The compound was characterized using scanning electron microscopy (SEM) and X-ray diffraction (XRD). It was also evaluated the silver ions concentration in the compound through an ion-specific electrode. The antimicrobial action in planktonic cells was evaluated by minimum inhibitory concentration (MIC) and minimum bactericidal or fungicidal concentration (MBC / MFC). SEM analyzes showed Ag-NP decorating the surface of the TMP particles and the effectiveness of the reaction was demonstrated by the reduction of silver ions greater than 90%. Both compounds presented antimicrobial activity against the evaluated microorganisms, however, Candida albicans was more susceptible. For Streptococcus mutans the MIC and MBC values were 5850 μg / ml and 11700 μg / ml respectively for compound T1, and 1463 μg / ml and 5850 μg / ml for compound T10. Thus, compounds synthesized by Ag-NP...A fim de se desenvolver novos agentes antimicrobianos, muitas pesquisas vêm sendo conduzidas, e dentre estes agentes podemos citar as nanopartículas de prata (NP-Ag), devido a sua atividade antimicrobiana e antifúngica contra um amplo espectro de microorganismos. Além disso, o trimetafosfato de sódio (TMP) preserva a integridade e estabilidade do esmalte e sua associação nas proporções adequadas com o fluoreto (F) pode proporcionar um efeito protetor maior em relação aos produtos fluoretados convencionais. Assim, objetivo do presente estudo foi sintetizar um composto contendo nanopartículas de prata (NP-Ag), trimetafosfato de sódio (TMP) e fluoreto (F), caracterizá-lo e avaliar a ação antimicrobiana contra células planctônicas de Candida albicans e Streptococcus mutans. Para isso, NP-Ag foram sintetizadas através da redução do nitrato de prata contendo 1 (T1) e 10% (T10) em relação ao TMP por meio de uma via química na presença de F e TMP. O composto foi caracterizado utilizando a microscopia eletrônica de varredura (MEV) e difração de raios-X (DRX). Foi avaliada também a quantidade de íons prata no composto através de eletrodo íon-específico. A ação antimicrobiana em células planctônicas foi avaliada através da concentração inibitória mínima (CIM) e concentração bactericida ou fungicida mínima (CBM/CFM). As análises de MEV apresentaram NP-Ag decorando a superfície das partículas de TMP para os compostos sintetizados e a efetividade das reações foi demonstrada pela redução de íons prata maior que 90%. Ambos os compostos apresentaram atividade antimicrobiana contra os microrganismos avaliados, sendo a Candida albicans mais susceptível.
