A ação da genisteína no reparo ósseo alveolar e remodelação do colo do fêmur de ratas ovariectomizadas

Resumo: In mandibular and maxillary bones, hypoestrogenism affects the alveolar repair process, reducing bone formation after tooth extraction. And in the process of remodeling the bone tissue provides progressive decrease of bone mass. Among the drugs studied in the attempt to minimize these effects, soy isoflavones, such as genistein (Gen), are considered natural modulators of the estrogen receptor because of their conformational binding. However, its action on bone tissue remains controversial. The objective of this study was to analyze the action of Gen in alveolar bone repair and remodeling of the femoral neck in ovariectomized rats (OVX). Fourty Wistar rats (6 months) were randomly assigned to the following groups: Sham/NaCl; Sham/Gen; OVX/NaCl; OVX/Gen. The vaginal smear of the animals was collected daily to verify the estrous cycle for 15 days and only animals with regular estrous cycle were accepted for our study. Subsequently, the rats of the OVX groups at the 60 month were OVX, and the rats of the Sham groups underwent the ovarian removal mock surgery. Ten days after surgeries, the Sham/Gen and OVX/Gen experimental groups received 5.76 mg/kg/day of Gen, and the Sham/NaCl and OVX/NaCl groups received 0.3 mL of NaCl by gavage during 90 days. The right upper central incisor was extracted after 62 days of substitutive therapy. At the end of the 90 days, they were sacrificed, the right alveolus was removed and the left femur was collected. The alveolar middle third region was analyzed by histometric analysis and immunohistochemical analysis of protein immunolabeling, Tartarate Resistant Acid Phosphatase (TRAP) and Osteocalcin (OCN). E no processo de remodelação do tecido ósseo proporciona progressiva diminuição da massa óssea. Entre os fármacos estudados na tentativa de minimizar esses efeitos, tem as isoflavonas derivadas da soja, como a genisteína (Gen), consideradas modulador natural do receptor de estrógeno, devido sua ligação conformacional. Entretanto, sua ação sobre o tecido ósseo permanece controvérsia. O objetivo deste estudo foi analisar a ação da Gen no reparo do osso alveolar e processo de remodelação do colo de fêmur em ratas ovariectomizadas (OVX). Foram utilizadas 40 ratas da linhagem Wistar (6 meses), distribuídas aleatoriamente nos grupos: Sham/NaCl; Sham/Gen; OVX/NaCl; OVX/Gen. O esfregaço vaginal dos animais foi colhido diariamente para verificação do ciclo estral, durante 15 dias e foram aceitos somente animais com ciclo estral regular para nosso estudo. Posteriormente, as ratas dos grupos OVX, no 60 mês foram OVX, e as ratas dos grupos Sham foram submetidas à cirurgia fictícia de remoção dos ovários. Dez dias após as cirurgias, os grupos experimentais Sham/Gen e OVX/Gen receberam 5,76 mg/kg/dia de Gen, e os grupos Sham/NaCl e OVX/NaCl receberam 0,3 mL de NaCl, por gavagem, durante 90 dias. Foi realizada extração do incisivo central superior direito após 62 dias de terapia substitutiva. No final dos 90 dias, foram sacrificados, fez-se a separação do alvéolo direito e a coleta do fêmur esquerdo. A região do terço médio alveolar foi analisada por análise histométrica e análise imunoistoquímica das imunomarcações das proteínas, Fosfatase Ácida Resistente ao Tartarato (TRAP) e Osteocalcina (OCN).
