Análise proteômica do tecido hepático de camundongos com diabetes mellitus induzidos por estreptozotocina e dieta hiperlipídica

Resumo: Diabetes mellitus affects millions of people, and can cause serious complications such as heart attacks, strokes, kidney failure, among others. Proteomics research provides a clearer metabolic view of mechanistic processes that reveal new ways of treatments, preventions and diagnosing. Diabetes mellitus is a complex and controversial disease. Many metabolic pathways, biological processes and protein alterations are not completely understood and can vary among researches, hence is essential the use of even more precise methods for its analysis. Thus the objective of this study was to evaluate how hepatic protein expression changes, through proteomic analysis of the functioning of mice with or type 2 diabetes mellitus. Thereunto, 18 mice were divided into 3 groups (n=6). The first is formed by animals that received standard diet and SZT injections (CD1), the second are animals that received high fat diet and STZ injections (CD2), and the third group received standard diet (CND), the latter is the control group. The model used was successfully implanted and revealed significant changes in the metabolism of DM2, related to the metabolism of carbohydrates, lipids and proteins. The high-fat diet proved to be a potentiator of the adverse effects of DM2 on the liver. This project also proposed new targets for the understanding of this pathology.

As análises proteômicas fornecem uma visão metabólica mais clara dos processos mecanísticos por trás das patologias analisadas permitindo revelar novas formas de diagnósticos, tratamentos e prevenções. O diabetes mellitus é uma doença complexa, e controversa. Muitas das vias metabólicas, processos biológicos e proteínas alteradas ainda não são compreendidas e variam entre as pesquisas, desse modo é essencial a utilização de métodos mais sensíveis para sua análise. O objetivo deste trabalho foi avaliar as alterações de expressão das proteínas hepáticas, por meio da análise proteômica do fígado de camundongos com ou sem diabetes mellitus tipo 2. Para isso foram utilizados 18 camundongos divididos em 3 grupos (n=6). O primeiro foi de animais que receberam a dieta padrão e injeções de SZT (CD1), o segundo foram animais que receberam dieta hiperlipídica e injeções de SZT (CD2) e em terceiro, o grupo que recebeu dieta padrão (CND), este último é o controle. O modelo utilizado foi implantado com sucesso e revelou alterações significativas no metabolismo do DM2, relacionados ao metabolismo de carboidratos, lipídios e proteínas. A dieta hiperlipídica se mostrou uma potencializadora dos efeitos adversos do DM2 no fígado.
