Resumo: Currently, it is popular knowledge that natural products can provide more health benefits than synthetic chemicals. An example of this is what can be seen with respect to bee venom, which is a bee product and has been widely used, long ago, in traditional Oriental medicine for the treatment of arthritis and pain. However, recently, bee venom has also been used on cosmetic industries due to its potential effect on the skin on reducing wrinkles. Among compounds from bee venom, melittin is its major constituent. This use of the bee venom is given by the melittin characteristic of inducing an increase on blood circulation and, consequently, acts on the production of collagen and elastin, removing dead cells and reducing wrinkles. Nowadays, it is known that it acts on healthy cells, disrupting them, once melittin binds on plasmatic membrane forming water pores or increasing cell permeability. Thus, the present study aimed to evaluate the cytotoxic, genotoxic and mutagenic potentials of melittin in human hepatocarcinoma cells (HepG2). In this study, MTT test, Trypan blue and oxidative stress assays (cytotoxicity); the comet assay (genotoxicity) and the micronucleus with cytokinesis block assay (mutagenicity) were performed. For the MTT assay (cytotoxicity test) with melittin the following concentrations were tested: 5.0 μg/mL, 1.0 μg/mL, 0.5 μg/mL, 0.05 μg/mL, 0.005 μg/mL, 0.0005 μg/mL. Of these, only the concentration of 5.0 μg/mL was cytotoxic to the cells. The other concentrations (1.0 μg/mL, 0.5 μg/mL and 0.05 μg/mL) had a cell viability higher than 80%, so they were used on comet, MN and oxidative stress assays. In the Comet assay, none of them showed genotoxic potential to cells, which allows to conclude that these concentrations were not able to induce damage or, if there was damage induction, the cells were able to recover in the 24 hour period that they were exposed to melittin. When using the same concentrations on the MN...Atualmente, é de conhecimento popular que produtos naturais possam apresentar mais benefícios a saúde do que químicos sintéticos. Um exemplo disso é o que pode ser visto com respeito ao veneno de abelha, um produto apícola que vem sendo muito utilizado, há muito tempo, na medicina oriental tradicional para o tratamento de artrites e dores. Porém, recentemente, o veneno de abelha tem também sido utilizado também pela indústria de cosméticos, devido ao seu potencial efeito sobre a pele, na redução de rugas. Dentre os compostos do veneno de abelha, a melitina é a substância encontrada em maior quantidade. O uso do veneno como fármaco se deve as características da melitina, pois esta induz um aumento da circulação sanguínea e, consequentemente, atua na maior produção de colágeno e elastina, eliminando as células mortas e reduzindo as rugas. Hoje, sabe-se que a melitina também age sobre células saudáveis, causando seu rompimento, uma vez que se liga à membrana plasmática formando poros d'água ou aumentando a permeabilidade celular. Dessa forma, o presente estudo teve como objetivo avaliar o potencial citotóxico, genotóxico e mutagênico da melitina em células de hepatocarcinoma humano (HepG2). Neste estudo foram desenvolvidos ensaios do MTT, do Azul de Tripan e de estresse oxidativo (citotoxicidade); ensaio do cometa (genotoxicidade) e teste do micronúcleo com bloqueio da citocinese (mutagenicidade). Para o teste do MTT, realizado com a melitina, foram utilizadas as seguintes concentrações: 5,0 μg/mL, 1,0 μg/mL, 0,5 μg/mL, 0,05 μg/mL, 0,005 μg/mL, 0,0005 μg/mL. Destas concentrações, apenas a de 5,0 μg/ml foi citotóxica para as células. As demais concentrações (1,0 μg/mL, 0,5 μg/mL e 0,05 μg/mL) tiveram viabilidade celular maior que 80%, portanto utilizadas nos ensaios do cometa, MN e estresse oxidativo.
