Resumo: | Raphanus raphanistrum L. belongs to the family Brassicaceae, native to southern Europe widely disseminated across regions of temperate and subtropical world. Weed of economic importance in production fields poses threats to areas where wheat is sown for having suspected resistance to the ALS inhibitors. With the objective of confirm the resistance R. raphanistrum herbicide metsulfuron methyl evaluate the existence of cross-resistance and multiple resistance and meet the competitive ability of the weed, we conducted experiments in a greenhouse at the University of Passo Fundo, in the year 2011/12. To confirm the resistance R. raphanistrum metsulfurom methyl herbicide, we conducted experiments in which the treatments consisted of the use of resistant biotypes (biotype R) and susceptible (biotype S) and ten increasing doses of the herbicide. We evaluated control, reduced size and dry matter (DM). Regression was using nonlinear adjustment of the dose response curve. The biotype R was resistant to the herbicide metsulfuron methyl. To evaluate the existence of cross-resistance and multiple resistance, as well as control of the biotype R was held experiment where the treatments consisted of biotype R and biotype S, 12 herbicides (glyphosate, atrazine, bentazone, 2,4-D, sulfometuron methyl + chlorimuron ethyl, chlorimuron ethyl, cloransulam methyl, iodosulfuron, nicossulfuron, imazethapyr, metsulfuron methyl and Imazapic + imazapyr) and untreated control. Evaluated control and DM. The biotype R has cross-resistance to chemical group’s imidazolinones, sulfonylureas and triazolopyrimidines. The biotype R showed no multiple resistance. The herbicide glyphosate, bentazone, atrazine and 2,4-D shown to be effective in the control of biotype R. To assess the competitive ability of the biotypes Rand biotype S in relation to the culture of wheat, were conducted three experiments arranged in replacement series: (1 - wheat with the biotype R; 2 - wheat with the biotype S; and 3 – the biotype R with the biotype S) at the following ratios: 100:0, 75:25, 50:50, 25:75, and 0:100. Analyzed the competitiveness through diagrams applied to replacement and competitiveness indices with evaluation of the dry matter of shoots and area leaf. The biotype R and biotype S demonstrated superior competitive ability of wheat. The competitiveness of the biotype R compared to the biotype S was similar. |

Raphanus raphanistrum L. e Raphanus sativus L., também conhecidas por nabo ou nabiça, são espécies cosmopolitas invasoras em diversas culturas agrícolas, causando prejuízos principalmente em lavouras de trigo, devido ter evoluído resistência aos herbicidas inibidores da acetolactato sintase (ALS). A ALS é enzima chave envolvida na biossíntese de aminoácidos valina, leucina e isoleucina. A inibição da enzima ALS, causada pela ação de herbicidas, afeta a síntese de aminoácidos e, consequentemente, leva a morte da planta. Desvendar o mecanismo de resistência aos herbicidas inibidores da ALS, em R. raphanistrum e R. sativus é importante para manejo racional da resistência e adoção de futuras tecnologias direcionadas a tal fim. Com o objetivo de elucidar o mecanismo de resistência e avaliar a herança da resistência em populações de R. raphanistrum e R. sativus, aos herbicidas inibidores da ALS, realizou-se pesquisas no Brasil e Canadá. Populações de R. raphanistrum e R. sativus oriundas de lavouras de trigo, do estado do Paraná e do Rio Grande do Sul, foram conduzidas em casa de vegetação e através de autopolinizações, levados a homozigose para a resistência. Plantas de três a quatro folhas, oriundas de sementes homozigotas, foram selecionadas para extração do DNA genômico e extração da enzima ALS. A partir do DNA extraído, realizou-se a amplificação do gene ALS. Os fragmentos do gene ALS foram purificados, sequenciados e comparados com sequências de aminoácidos de R. raphanistrum já conhecidas na literatura. Também se realizou o teste de dose resposta em todas as populações de Raphanus spp., utilizando o herbicida metsulfurom-metil in vivo. Para determinar os parâmetros enzimáticos da enzima ALS in vitro, fez-se a extração da enzima e avaliou-se sua atividade na presença do herbicida imazapic e, a concentração de substrato (piruvato) que fornece velocidade inicial igual à metade da velocidade máxima de reação (KM) e a velocidade máxima de reação (Vmáx). Utilizando plantas de três a quatro folhas, avaliou-se a presença ou não de resistência metabólica, pelo complexo citocromo P450 monooxigenase, através da utilização do herbicida metsulfurom-metil antecedido da aplicação de malation, sendo este último, inibidor da P450. Em experimento para avaliar herbicidas alternativos no controle de Raphanus spp. resistentes aos herbicidas inibidores da ALS e, possível resistência cruzada ou resistência múltipla, foram aplicados vários herbicidas, pertencentes a diferentes grupos químicos dos inibidores da ALS e herbicidas pertencentes a outros mecanismos de ação. A herança da resistência foi avaliada através de cruzamentos entre populações resistentes e suscetíveis, dentro de cada espécie. Utilizou-se o herbicida metsulfurommetil como agente selecionador para o F1 resistente ou suscetível oriundo dos cruzamentos. O mecanismo de resistência em R. raphanistrum e R. sativus foi devido a mutação Trp-574-Leu no gene ALS. A dose resposta ao herbicida metsulfurom-metil confirmou a insensibilidade a este herbicida nas populações resistentes em estudo. As populações resistentes de R. raphanistrum e R. sativus demonstraram insensibilidade ao herbicida imazapic. Houve alteração nos parâmetros cinéticos da enzima ALS através do aumento no KM e Vmáx nas populações resistentes comparadas as populações suscetíveis, nas duas espécies. Não há envolvimento do complexo citocromo P450 monooxigenase na resistência de R. raphanistrum e R. sativus aos herbicidas inibidores da ALS. R. raphanistrum e R. sativus possuem resistência cruzada aos herbicidas pertencentes aos grupos químicos sulfonilureias, imidazolinonas e triazolopirimidinas. Os herbicidas atrazina, 2,4-D, glifosato, saflufenacil, bentazona, flumioxazina, glufosinato de amônia e tembotriona são alternativas para o manejo da resistência nessas populações.
