Resumo: The effect of the treatment of the polimeric film preparation with the extracted polyssacharíd of the gum of the Anacardium occidentale L. was evaluated in excisionals cutaneous injuries in mice albinic Swiss, through the accompaniment of the evolution of the healing cutaneous process, under the clinical and histopathological aspects. After the accomplishment of the surgical procedure, 5 groups (n=15/grupo) had been formed in accordance with the employed treatment: 1. NaCl (NaCl 0,15M); 2. AA (acetic acid 4% v/v); 3. POLICAJU/AA (polimeric film I containig acetic POLICAJU 10% m/v in acid 4% v/v) and 4, POLICAJU/NaCl (polimeric film I containing POLICAJU 10% m/v in NaCl 0,15M). Tissue samples had been fixed and staining (Tricrômicro de Masson) and analyzed in 2nd, 7th and 12th pos-surgical (P.S.) days. In group POLICAJU/AA it was observed reduction of the flogistics signals represented by presence of edema, crust and exudate. Histopathological point of view, the groups treatment with POLICAJU had presented partial reepitelization and soft granulation tissue, specially in the POLICAJU/NaCl group. Thus, it is concluded that the polímeric films I containing POLICAJU, applied as oclusive dressings, had been efficient in the process of cutaneous tecidual repair. The use of films obtained of the POLICAJU for tripsina immobilization (DEI), by entrepment, and its application as dressing in cutaneous injuries was evaluated. DEI it demonstrated it to a constant release of the trypsin optimal pH and temperature of 8,6 and 55°C, in the analyzed conditions and revealed stability stored in the conditions retained about 80% (28 days 25°C) of its initial activity. The wound treatment withDEI it had more presented organized morphologic standard when compared with the controls (NaCl 0,15M and acetic acid 4% v/v). These results demonstrate that the POLICAJU can be used as matrix for trypsin immobilization and a biomaterial potential. An experimental design (2) was applied for determination of the best conditions of entrapment of the trypsin in POLICAJU films. For this study the effect of the concentration of trypsin and the temperature on its activity had been analyzed. It was observed that of the analyzed parameters the lesser concentration of trypsin (100ug/mL) was the best condition of immobilization. This system was evaluated how much it influences enviromental (pH and temperature) and topic application. The best values of pH and the temperature had been 9,0 and 55C, demonstrating to stability the range of temperature (25-65C), how much to the storage DEI it retained 30% of its activity (28 days). The results of the hystopathological evaluations had demonstrated an acceleration of the healing process. From the results of the immobilization of trypsin entrappment in POLICAJU/NaCl and POLICAJU/AA, DEI it was applied as oclusive dressing in the treatment of cutaneous wounds being evaluated its effect under the clinical aspects and histopathology. After the surgical procedure 5 groups had been formed in accordance with the type of treatment (n=15/grupo): 1. NaCl; 2. AA; 3. AAT; 4. POLICAJUT/AA and 5, POLICAJUT/NaCl. Tissue samples had been fixed and staining(Tricrômicro of Masson) in the 2nd, 7th and 12th days after the surgery. The wound treatment with POLICAJUT/NaCl how much to the clinical and histopathology parameters they had presented one better when comparative evolution to the too much groups. The results gotten from all the studies of immobilization and biological application demonstrate the effectiveness of the POLICAJUT/NaCl film with oclusive dressing.

Após a realização do procedimento cirúrgico, 4 grupos (n=15/grupo) foram formados de acordo com o tratamento empregado: 1, NaCl (NaCl 0,15M); 2, AA (ácido acético 4% v/v); 3, POLICAJU/AA (filme polimérico contendo POLICAJU 10% m/v em ácido acético 4% v/v) e 4, POLICAJU/NaCl (filme polimérico contendo POLICAJU 10% m/v em NaCl 0,15M). Amostras de tecido foram fixadas e coradas (Tricrômicro de Masson) e analisada no 2o, 7o e 12o dias P.O.. No grupo POLICAJU/AA foi observado diminuição dos sinais flogísticos representados por presença de edema, crosta e exsudato. Do ponto de vista histopatológico, os grupos tratados com POLICAJU apresentaram reepitelização parcial e tecido de granulação fibroso, especialmente no grupo POLICAJU/NaCl. Assim, conclui-se que os filmes polímericos contendo POLICAJU, aplicados como curativos oclusivos, foram eficazes no processo de reparo tecidual cutâneo. A utilização de filmes obtidos do POLICAJU como matriz para imobilização de tripsina (DEI), por enclausuramento, e sua aplicação como curativo em lesões cutâneas foi avaliada. O DEI demonstrou uma liberação constante da tripsina, pH e temperatura ótimos de 8,6 e 55°C, nas condições analisadas e mostrou-se estável nas condições do meio, retendo cerca de 80% (28 dias a 25°C) da sua atividade inicial. As lesões tratadas com o DEI apresentaram padrão morfológico mais organizado quando comparado aos controles (NaCl 0,15M e ácido acético 4% v/v). Estes resultados demonstram que o POLICAJU pode ser utilizado como matriz para imobilização de tripsina e um potencial biomaterial. Um planejamento experimental (22) foi aplicado para determinação das melhores condições de enclausuramento da tripsina em filmes de POLICAJU. Para esse estudo foram analisados os efeitos da concentração de tripsina e da temperatura sobre sua atividade. Observou-se que dos parâmetros analisados a menor concentração de tripsina (100μg/mL) foi a melhor condição de imobilização. Este sistema foi avaliado quanto a influência do meio (pH e temperatura) e aplicação tópica. Os valores ótimos de pH e a temperatura foram 9,0 e 55oC, demonstrando estabilidade a ampla faixa de temperatura (25-65oC), quanto ao armazenamento o DEI reteve cerca de 30% de sua atividade (28 dias). Os resultados das avaliações histopatológicas demonstraram uma aceleração do processo cicatricial. A partir dos resultados da imobilização de tripsina enclausurada em POLICAJU/NaCl e POLICAJU/AA, o DEI foi aplicado como curativo oclusivo no tratamento de lesões cutâneas sendo avaliado seus efeitos sob os aspectos clínicos e histopatológico. Após o procedimento cirúrgico 5 grupos foram formados de acordo com o tipo de tratamento (n=15/grupo): 1, NaCl; 2, AA; 3, AAT; 4, POLICAJUT/AA e 5, POLICAJUT/NaCl. Amostras de tecido foram fixadas e coradas (Tricrômicro de Masson) nos 2o, 7o e 12o dias após a cirurgia. As lesões tratadas com POLICAJUT/NaCl quanto aos parâmetros clínicos e histopatológicos apresentaram uma melhor evolução quando comparado aos demais grupos. After the accomplishment of the surgical procedure, 5 groups (n=15/grupo) had been formed in accordance with the employed treatment: 1. NaCl (NaCl 0,15M); 2. AA (acetic acid 4% v/v); 3. POLICAJU/AA (polimeric film I containig acetic POLICAJU 10% m/v in acid 4% v/v) and 4, POLICAJU/NaCl (polimeric film I containing POLICAJU 10% m/v in NaCl 0,15M). Tissue samples had been fixed and staining (Tricrômicro de Masson) and analyzed in 2nd, 7th and 12th pos-surgical (P.S.) days. In group POLICAJU/AA it was observed reduction of the flogistics signals represented by presence of edema, crust and exudate. Histopathological point of view, the groups treatment with POLICAJU had presented partial reepitelization and soft granulation tissue, specially in the POLICAJU/NaCl group. Thus, it is concluded that the polímeric films I containing POLICAJU, applied as oclusive dressings, had been efficient in the process of cutaneous tecidual repair. The use of films obtained of the POLICAJU for tripsina immobilization (DEI), by entrepment, and its application as dressing in cutaneous injuries was evaluated. DEI it demonstrated it to a constant release of the trypsin optimal pH and temperature of 8,6 and 55°C, in the analyzed conditions and revealed stability stored in the conditions retained about 80% (28 days 25°C) of its initial activity. The wound treatment withDEI it had more presented organized morphologic standard when compared with the controls (NaCl 0,15M and acetic acid 4% v/v). These results demonstrate that the POLICAJU can be used as matrix for trypsin immobilization and a biomaterial potential. An experimental design (2) was applied for determination of the best conditions of entrapment of the trypsin in POLICAJU films. For this study the effect of the concentration of trypsin and the temperature on its activity had been analyzed. It was observed that of the analyzed parameters the lesser concentration of trypsin (100ug/mL) was the best condition of immobilization. This system was evaluated how much it influences enviromental (pH and temperature) and topic application. The best values of pH and the temperature had been 9,0 and 55C, demonstrating to stability the range of temperature (25-65C), how much to the storage DEI it retained 30% of its activity (28 days). The results of the hystopathological evaluations had demonstrated an acceleration of the healing process. From the results of the immobilization of trypsin entrappment in POLICAJU/NaCl and POLICAJU/AA, DEI it was applied as oclusive dressing in the treatment of cutaneous wounds being evaluated its effect under the clinical aspects and histopathology. After the surgical procedure 5 groups had been formed in accordance with the type of treatment (n=15/grupo): 1. NaCl; 2. AA; 3. AAT; 4. POLICAJUT/AA and 5, POLICAJUT/NaCl. Tissue samples had been fixed and staining(Tricrômicro of Masson) in the 2nd, 7th and 12th days after the surgery. The wound treatment with POLICAJUT/NaCl how much to the clinical and histopathology parameters they had presented one better when comparative evolution to the too much groups. The results gotten from all the studies of immobilization and biological application demonstrate the effectiveness of the POLICAJUT/NaCl film with oclusive dressing.
