Resumo: The oral mucosa consists of highly proliferative squamous epithelium that under normal conditions performs its functions in a harmonic way with the underlying tissues. Bisphosphonates are synthetic, anti-resorptive drugs used for the control and treatment of diseases of bone follow-up, such as: Paget's diseases; metastatic bone tumors; osteopenias and osteoporosis. This drug acts inhibiting enzymes major for proliferation and migrating cellphone, monitor for the effects of cellular phones of hard and soft. The present death is evaluated by the effects of zoledronic acid (ZL) on the epithelium and the lamina propria of the palatal mucosa of rats chronically accompanied with zoledronic acid by intravenous route. Twenty-four male Wistar rats with a mean of 20 months, weighing about 500 grams, were used. In all groups, CT (Control) - received physiological saline 0.9% (SF) via EV, without volume of 0.1 ml; Group ZL - treated with zoledronic acid given via (EV) at a dose of 35 μg / kg every two weeks (Hokugo et al., 2010) and in four subgroups according to the number of doses: GCT-6 and GZL-6 (six doses), GCT-8 and CZL-8 (high doses). In these periods, the animals were submitted to euthanasia to allow the removal of the maxilla of staining with the aid of tests for PCNA to allow qualitative and quantitative quantitative analysis. Results do not indicate differences in cell thickness. Changes in the morphology and pattern of the basal layer, such as hypercorradant cells, increased melanocyte presence, increased keratohyaline formation in the granulomatous cells and interface of the granulosa layer with the cornea were found in the experimental group (ZL). Decrease of cells immunolabeling to PCNA+ has been found between GCT-08 to GZL-08, showing citotoxic effect of the Zoledronic Acid in this cell lineage. When it comes to tissues in the bone tissue and it is suggested to investigate, concomitantly, as changes in the underlying bone tissue.

Os bisfosfonatos são drogas sintéticas, antirreabsortivas, utilizados para controle e tratamento de doenças de acometimento ósseo e atuam inibindo enzimas da via do Mevalonato fundamentais para proliferação e migração celular, afetando linhagens celulares de tecidos duros e moles. O objetivo deste trabalho foi avaliar os efeitos do acido zoledrônico (ZL) sobre o epitélio e lamina própria da mucosa palatina de ratos tratados por 12 e 16 semanas com ácido zoledrônico por via endovenosa. Foram utilizados 24 ratos machos Albinos Wistar com idade média de 17 meses, pesando cerca de 500 gramas. Os animais foram divididos em dois grupos, CT (Controle) – receberam soro fisiológico 0,9% (SF) via EV, no volume de 0,1ml; Grupo ZL - tratados com o ZL administrado via (EV) na dose de 35 µg/Kg a cada duas semanas, resultando em quatro subgrupos de acordo com a quantidade de doses: GCT-6 e GZL-6 (seis doses), GCT-8 e GZL-8 (oito doses). Após os períodos determinados, os animais foram submetidos à eutanásia e removido as maxilas para processamento histotécnico de rotina para coloração com HE e imunohistoquímica para PCNA. Os resultados não apontaram diferenças na espessura das camadas celulares. Para ambos os grupos GCT-06 e GZL- 06 observou-se presença de melanócitos, queratinócitos volumosos na camada granulosa, grande formação de grânulos de queratohialina na camada granulosa e interface da camada córnea. No grupo GZL 06 ocorreu hipercromatismo de células basais, presença de células basais cubóides e acantose da camada espinhosa. Para os grupos GCT-08 e GZL-08 ambos apresentaram redução do número de camadas celulares, diminuição da formação dos grânulos de queratohialina, contudo, no grupo GZL-08 ocorreu à formação de cristas epiteliais alongadas sem alteração inflamatória celular. A imunomarcação de células PCNA+ manteve padrão discreto e homogêneo com exceção do grupo controle com 08 doses que apresentou maior marcação, seguido da diminuição da imunomarcação para o grupo experimental com 8 doses. As alterações encontradas podem estar relacionadas à: idade dos animais, protocolo de aplicação da droga, classe medicamentosa da droga, citotoxicidade e atuação do ZL.
