Produção e extração de carotenoides produzidos por Sporobolomyces ruberrimus utilizando subprodutos industriais

Resumo: | Biotechnological production using industrial byproducts in order to obtain products with high added value is the result of extensive research nowadays, aiming not only at the reduction of costs but also the decrease of environmental impacts. The purpose of this work was to study the kinetics of carotenoids growth and production by Sporobolomyces ruberrimus using cane and soy molasses as carbon source. Until now no study had been done with this yeast using these substrates. This study was divided into three parts. In the first part the characterization of carbon sources was made, in order to know the composition of byproducts available, and their subsequent use for the growth of yeast. In the second stage, preliminary studies were performed in Erlenmeyer flask, to verify the possibility of using industrial byproducts in the production of yeast. In addition to the microbial growth it was observed the yeast ability to accumulate carotenoids. In this stage it was observed the importance of oxygen availability in the culture medium and a low initial concentration of cells (1x106 cells / ml) for the proper development of the microorganism. The cane molasses showed the best results (μmáx = 0.06 h-1 , Xmax = 2.78 g.L -1 and Yx/s = 21 %) and was chosen to be used in the continuity of the proposed study. Initial studies of carotenoids extraction showed DMSO as the best solvent due to a higher efficiency compared to other solvents tested, the ability to permeate Sporobolomyces ruberrimus cell wall yeast and its low toxicity. For the recovery of carotenoids, after permeabilization of the yeast cell wall, ether was chosen for its optimal separation of the pigments and its low boiling point, which facilitates the carotenoids purification stage. The last stage was aimed at production, extraction and characterization of carotenoids extracted from yeast obtained in a bioreactor with cane molasses as a substrate, based on the results obtained in previous stages. The results obtained for the microbial growth kinetics and carotenoids production in the cultivation in bioreactor (μmáx = 0.1083 h-1 , Xmax = 14.8 gL-1 , Yx/s = 45 % and 549 µg of carotenoids/g.cel) confirmed the expectations of preliminary studies in Erlenmeyer flask. Although the results are preliminary, and non-optimized, the use of cane molasses as a substrate for production of carotenoids by the yeast Sporobolomyces ruberrimus, proved to be quite promising. |

Este trabalho teve por objetivo o estudo da cinética de crescimento e produção de carotenoides por Sporobolomyces ruberrimus, utilizando melaço de cana e melaço de soja como fonte de carbono. Até o momento nenhum estudo havia sido feito com esta levedura utilizando estes substratos. Este estudo foi dividido em três partes. Na primeira parte foi feita a caracterização das fontes de carbono usadas, com o objetivo de conhecer a composição dos subprodutos disponíveis, e sua posterior utilização para o crescimento da levedura. Na segunda etapa, foram realizados estudos preliminares em Frascos de Erlenmeyer, para verificar a possibilidade da utilização dos subprodutos industriais na produção da levedura. Além do crescimento microbiano, foi observada a capacidade da levedura de acumular carotenoides. Nesta etapa foi observada a importância da disponibilidade de oxigênio no meio de cultura e de uma baixa concentração inicial de células (1x106 células/mL), para o desenvolvimento adequado do microrganismo. O melaço de cana apresentou os melhores resultados (µmáx = 0,06 h -1 , Xmáx = 2,78 g.L -1 e YX/S = 21 %) e foi escolhido para ser utilizado na continuidade do estudo proposto. Os estudos iniciais de extração de carotenoides apontaram o DMSO como melhor solvente, pela maior eficiência, em relação aos outros solventes testados, na capacidade de permear a parede das células da levedura Sporobolomyces ruberrimus e por sua baixa toxidade. Para a recuperação dos carotenoides, após a permeabilização da parede celular da levedura, o éter foi escolhido pela ótima separação dos pigmentos e pelo seu baixo ponto de ebulição, o que facilita a etapa de purificação dos carotenoides. A última etapa foi destinada a produção, extração e caracterização dos carotenoides extraídos da levedura obtida em biorreator com melaço de cana como substrato, baseados nos resultados obtidos nas etapas anteriores. Os resultados obtidos para a cinética de crescimento microbiano e para a produção de carotenoides no cultivo em biorreator (µmáx = 0,1083 h -1 , Xmáx = 14,8 gL-1 , YX/S= 45 % e 549 µg de carotenoides/g.cel), confirmaram as expectativas dos estudos preliminares em Frasco de Erlenmeyer.
