Integrative review: verification of efficacy / effectiveness of medical Cannabis and cannabinoid derivatives in Alzheimer´s disease

Resumo: Considering the limited therapeutic effects of current medications for trying to treat or reduce the symptoms of Alzheimer's disease, it is relevant to search for new therapeutic alternatives, with significant efficacy and reduced side effects. One of the promising solutions to prevent the progression of behavioral and cognitive changes in the disease are cannabinoid derivatives, in which in vivo studies have shown a reduction in oxidative stress, neuroinflammation, formation of amyloid plaques and neurofibrillary tangles, in addition being related to regulation activation of microglial cells and release of macromolecules, all factors that, when present, contribute to the worsening and evolution of the disease. Medicinal Cannabis and cannabinoid derivatives such as cannabidiol and Δ9-tetrahydrocannabinol have shown very significant therapeutic efficacy for various diseases and symptoms, such as chronic pain, nausea and vomiting induced by chemotherapy, multiple sclerosis, anorexia nervosa, anxiety, Huntington's disease, Parkinson's disease, epilepsy, Alzheimer's disease, among others. Therefore, this work aimed to gather studies that would enable the analysis of the efficacy/effectiveness of cannabinoid derivatives in Alzheimer's disease in more updated publications.

Una de las soluciones prometedoras para prevenir la progresión de cambios conductuales y cognitivos en la enfermedad son los derivados cannabinoides, en los que estudios in vivo han demostrado una reducción del estrés oxidativo, neuroinflamación, formación de placas amiloides y ovillos neurofibrilares, además de estar relacionados con regulación de la activación de las células microgliales y liberación de macromoléculas, factores todos que, cuando están presentes, contribuyen al agravamiento y evolución de la enfermedad. El cannabis medicinal y los derivados cannabinoides como el cannabidiol y el Δ9-tetrahidrocannabinol han demostrado una eficacia terapéutica muy significativa para diversas enfermedades y síntomas, como dolor crónico, náuseas y vómitos inducidos por quimioterapia, esclerosis múltiple, anorexia nerviosa, ansiedad, enfermedad de Huntington, enfermedad de Parkinson, epilepsia, enfermedad de Alzheimer, entre otras. Por tanto, este trabajo tuvo como objetivo recopilar estudios que permitieran analizar la eficacia / efectividad de los derivados cannabinoides en la enfermedad de Alzheimer en publicaciones más actualizadas.   Considerando os efeitos terapêuticos limitados das medicações atuais para a tentativa de tratamento ou diminuição dos sintomas da doença de Alzheimer, torna-se relevante a busca por novas alternativas terapêuticas, com eficácia significativa e efeitos colaterais diminuídos. Uma das soluções promissoras para impedir a progressão das alterações comportamentais e cognitivas da doença são os derivados canabinoides, nos quais estudos in vivo têm mostrado uma redução no estresse oxidativo, neuroinflamação, formação de placas amiloides e emaranhados neurofibrilares, além de estar relacionado com a regulação da ativação das células da microglia e liberação de macromoléculas, todos os fatores que quando presentes, contribuem para a piora e evolução da doença. A Cannabis medicinal e os derivados canabinoides como o canabidiol e o Δ9-tetraidrocanabinol têm mostrado eficácia terapêutica bastante significativa para variadas doenças e sintomas, como dor crônica, náusea e vômito induzidos pela quimioterapia, esclerose múltipla, anorexia nervosa, ansiedade, doença de Huntington, doença de Parkinson, epilepsia, doença de Alzheimer, entre outros.
