Avaliação da resposta tecidual a discos de ácido poli-l/d láctico: estudo experimental em ovelhas

Resumo: The aim of this study was to evaluate the inflammatory reaction, more specifically the foreign body reaction and behavior of bone remodeling in sheep’s tibiae and ulnae after installation of copolymer discs consisting of 95% poly-L lactic acid and 5% acid poly-D Lactic (PLDLA). It was used two adult female sheep which was fixed, by screws, for circular discs of PLDLA in each tibia and ulna of the sheep. A total of 16 discs were fixed in each animal. The sheeps were randomly divided into two groups according to the time of death after surgery: 30 and 60 days. Subsequently the samples were processed, resulting in a slide for each sample. After that the slides were investigated in optical microscope with magnification of 40 x and 100 x. In the slices of periosteal were evaluated the inflammatory infiltrate in a descriptive way in an increase of 40 X and quantitatively, by counting inflammatory cells in five random fields per sample in an optical microscope in an increase of 100 X. The inflammatory density was quantified according to the following scale: grade 0 - absence of inflammatory infiltrate, grade 1 - less than 30 cells per field, grade 2 - 30 to 100 cells per field and grade 3 - greater than 100 cells per field. In bone tissue it was analyzed two random fields per sample and if some bone resorption area appeared in the area of contact with the discs, this was described. In the end of the study, histological sections of the both groups showed tissue composed of organized fibroblasts and the inflammatory infiltrate varied from absent to mild. Talking about bone resorption, it had association with compression of the discs to the bone tissue, without evidence of bone resorption caused by the discs. At the end of the study verified biocompatibility, abscense of osteolisys at macroscopic view, disc structure undefformed and appropriated fit between titanium screw and PLDLA disc.

Foram utilizadas duas ovelhas fêmeas adultas em que se fixaram, através de parafusos, quatro discos circulares de PLDLA em cada tíbia e ulna das ovelhas, totalizando 16 discos em cada animal. As ovelhas foram aleatoriamente divididas em dois grupos, de acordo com o período de morte após a cirurgia: 30 e 60 dias. Posteriormente as amostras foram processadas, obtendo-se uma lâmina por amostra, que foram investigadas ao microscópio óptico com aumentos de 40x e 100x. Nas lâminas de periósteo avaliou-se o infiltrado inflamatório de forma descritiva, em um aumento de 40 X, e de forma quantitativa, pela contagem de células inflamatórias em cinco campos aleatórios por amostra no microscópio óptico, em aumento de 100 X. A densidade inflamatória foi quantificada conforme a seguinte escala: grau 0 – ausência de infiltrado inflamatório, grau 1 – menor que 15 células por campo, grau 2 – 15 a 50 células por campo e grau 3 – maior que 50 células por campo. O tecido ósseo foi analisado macroscopicamente, e havendo área de reabsorção óssea na área de contato com os discos, esta foi descrita. Observou-se ao final do estudo que as secções histológicas dos grupos 30 de 60 dias apresentavam tecido conjuntivo composto por fibroblastos maduros, organizados e o infiltrado inflamatório variava de ausente a leve. Em relação à reabsorção óssea há associação com a compressão dos discos ao tecido ósseo, não havendo indícios de reabsorção óssea causadas pelos discos. No final do estudo verifica-se biocompatibilidade, ausência de osteólise a nível macroscópico, manutenção da estrutura do disco e adequada associação entre o parafuso de titânio e disco de PLDLA.Made available in DSpace on 2011-12-27T14:14:09Z (GMT).
