Resumo: Meat is widely consumed around the world. Besides, this food has a significant influence on the Brazilian economy. Regarding the nutritional terms, it is considered as a source of essential elements (e.g., molybdenum, copper, and manganese) that are important to the human organism. In this context, this thesis's goal is the development of analytical methods and the investigation of strategies for the determination of essential and toxic elements in meat by microwave-induced plasma optical emission spectrometry (MIP OES) and inductively coupled plasma – Tandem mass spectrometry (ICP-MS/MS). The use of MIP OES was investigated for the determination of phosphorus from phospholipids in meat. Due to the introduction of organic solvent, some strategies were adopted, such as the employment of an external gas control module (EGCM) and the signal correction applying internal standardization (Te) and molecular species (OH). The accuracy was estimated by comparing the results obtained with ion chromatography. The recovery found using the internal standardization was 95%, while molecular specie was 94%. These results indicated that these signal corrections were effective, and their employment significantly improved accuracy. The dispersive liquid-liquid microextraction (DLLME) was used to enhance the procedure’s sensitivity for the determination of molybdenum in meat by MIP OES. In this study, the selection of extracting and dispersing solvents was based on the mixture design. Additionally, the DLLME parameters were optimized by applying a Box-Behnken design. A bovine liver certified reference material (1577c, NIST) was employed to check the accuracy and a recovery of 92% was obtained. The enhancement factor and the limit of detection estimated were 4.5 and 0.20 µg g-1, respectively. In another work, a critical evaluation of nebulizer effects in a nitrogen plasma was performed. Four nebulizers (concentric, MiraMist, OneNeb series 1 and 2) were investigated concerning the plasma fundamental parameters (e.g., temperature and number of electron density) and solvent transport efficiency. Besides, the reproducibility was evaluated in different matrices, and the accuracy was investigated by analyzing four CRMs and s spiked sample from sugarcane spirit due to its alcoholic composition. The results demonstrated that the plasma fundamental parameters were considerably affected by the nebulizer type. Besides, the OneNeb series 2 showed the best performance considering the sensitivity, solvent transport efficiency and precision. The determination of nine essential and toxic elements (As, Cd, Co, Cr, Mn, Mo, Pb, Sr, and V) in meat samples was performed by ICP-MS/MS. Initially, the use of gases (O2, H2, and He) in the reaction octopolar system (ORS) was explored to minimize the spectral interferences. Adicionalmente, esse alimento possui uma grande influência na economia brasileira. Em termos nutricionais, a carne é considerada como fonte de elementos essenciais, como cobre, ferro, fósforo, manganês e molibdênio, e, que desempenham importantes funções no organismo humano. Nesse contexto, o objetivo dessa tese é o desenvolvimento de métodos analíticos e investigação de estratégias para a determinação de elementos essenciais e tóxicos em carne por espectrometria de emissão óptica com plasma induzido por micro-ondas (MIP OES) e espectrometria de massa com plasma acoplado indutivamente e configuração tandem (ICP-MS/MS). O uso do MIP OES foi investigado para a determinação do fósforo a partir dos fosfolipídios em carne. Devido a introdução de solvente orgânico no plasma, foram adotadas algumas estratégias como o uso do módulo de controle de gás externo (EGCM) e a correção do sinal do P por padronização interna (Te) e espécie molecular (OH). Para estimar a exatidão do método, os resultados foram comparados a valores obtidos por cromatografia iônica, e as recuperações foram 95% para a padronização interna e 94% para a espécie molecular. Os resultados demostraram que o uso dessas correções de sinais foram efetivas e representaram melhoria na exatidão. A microextração líquido-líquido dispersiva (DLLME) foi empregada para aumentar a sensibilidade de procedimento para a determinação de molibdênio em carne por MIP OES. As escolhas dos solventes dispersor e extrator foram baseadas nos planejamentos de misturas. Adicionalmente, os parâmetros da DLLME foram otimizados empregando um planejamento Box-Behnken. O material de referência certificado de fígado bovino (NIST, 1577c) foi analisado para avaliar a exatidão, sendo obtida recuperação de 92%. O fator de pré-concentração e o limite de detecção foram 4,5 e 0,20 µg g-1, respectivamente. Em outro estudo, foi realizada uma avaliação crítica dos nebulizadores para o plasma de nitrogênio. Neste tópico os efeitos de quatro nebulizadores (Concêntrico, MiraMist e OneNeb series 1 e 2) foram investigados quanto aos parâmetros fundamentais do plasma, como temperatura e densidade eletrônica. Adicionalmente foi avaliada a reprodutibilidade em diferentes matrizes. A exatidão foi investigada pela análise de quatro CRMs e pela adição e recuperação em amostra de cachaça, devido a seu teor alcoólico. Os resultados demostraram que os parâmetros fundamentais do plasma foram significativamente afetados pelos nebulizadores. Ademais, o nebulizador OneNeb serie 2 apresentou um melhor desempenho em relação a sensibilidade, eficiência de transporte e precisão. Em outro estudo, a determinação de nove elementos essenciais e tóxicos (As, Cd, Co, Cr, Mn, Mo, Pb, Sr e V) em amostras de carne foi realizada por ICP-MS/MS. Inicialmente, foi avaliado o uso dos gases (O2, H2 e He) no sistema octopolar de reação (ORS) para a minimização de possíveis interferências espectrais. A escolha da melhor condição operacional foi baseada na exatidão obtida durante a análise do material de referência de fígado bovino (NIST, 1577b). Para o Cd, Co, Mo, Pb e V não foi necessário o uso de gases. Por outro lado, para Cr e As, o uso do He foi mais adequado, enquanto o emprego do H2 apresentou melhor recuperação para o Sr. Os limites de detecção para esse método variaram entre 0,08 µg kg-1 (Cd) e 16 µg kg-1 (Mo). Posteriormente, amostras de carne bovina foram digeridas e analisadas em ICP-MS/MS sob condições ótimas.
