MICROPROPAGATION OF ERVA-MATE (Ilex paraguariensis St. Mil.)

Resumo: The objective of this study was to develop a micropropagation protocol for maté, which included the following experiments: I) Exam of culture medium contamination by microorganisms and in vitro development of explants; II) Pre-treatment of donor plants with the fungicides mancozeb and benomyl, amendment of ANA culture medium with various leveis of benomyl and rifampicin; III) Development of erva-mate explants in culture medium with different rates of cytokinin (BAP); IV) Mass propagation and root development of explants in two culture media (with agar + IBA or vermiculita + IBA). The fungi Alternaria sp. and Colletotrichum sp. were identified contam inating micropropagated explants and causing plant death. Bacteria were also found but they did not affect explant development. Pre-treatment of donor plants with fungicide reduced in vitro contamination by 90%. The use of 0.2 to 0.8 mg BAP in the culture medium did not stimulate shoot formation and did not influence leaf number. The rooting medium with IBA and vermiculita was more efficient on elongation and subsequent root emission.

Com o propósito de desenvolver um sistema para micropropagação da erva-mate, realizaram- se quatro experimentos: I) Observação de contaminantes fúngicos e bacterianos e deSenvolvimento de explantes cultivados in vitro; I I)Efeito da utilização de pré-tratamentos fúngicos (Mancozeb e Benomil) nas plantas matrizes e de combinações do fungicida (Benomil) e antibiótico (Rifampicina) no meio de cultura; III) Desenvolvimento dos explantes em meio de cultura, com diferentes dosagens de citocinina (BAP); IV) Subcultivo e enraizamento dos explantes em dois meios (ágar + IBA ou vermiculita + IBA). Inicialmente, identificaram-se os fungos Alternaria sp e Colletotrichum sp, que causaram a morte dos explantes; observou-se, também, a incidência de bactérias, as quais não interferiram no desenvolvimento dos mesmos. A utilização de pré-tratamentos fúngicos reduziu em 90% as contaminações iniciais ir, vitro. O uso de BAP (0,2 a 0,8 mg.L 1 ) no meio de cultura não estimulou a brotação e o número de folhas. O meio de enraizamento contendo IBA e vermiculita mostrou-se mais eficiente na elongação do material e emissão de raízes. Departamento de Diagnóstico e Pesquisa Agropecuária.
