Resumo: | The aim of this study was to evaluate in vitro the influence of two new formulations of the root canal sealers: Fillapex MTA (Angelus, Brazil) and BC Sealer (Brasseler, USA) and AH Plus (Dentsply, Germany) in cytotoxicity with cultured human cells. From bone fragments discarded after orthopedic surgery from healthy patients. Osteoblasts were cultured in the laboratory and, after confluence, were exposed to extracts of freshly mixed or set sealer for 24h. A group exposed to latex and a group of non-exposed cells served as positive and negative controls respectively. The cytotoxic effects of these sealers was performed by: a test multiparametric for cytotoxicity assay evaluating mitochondrial activity (XTT), membrane integrity (NR) and cell density (CVDE); test IC50; scanning electron microscopy; fluorescent dye with a test to vital cells and flow cytometry to the technology of using two Luminex ® panels involving human osteoclastogenesis. After statistical analysis, qualitative and morphological results showed that, compared with controls, the AH Plus was cytotoxic when it freshly (p <0.05) and when used set behavior was similar to the negative control. The MTA Fillapex was cytotoxic when freshly mixed or set, however, the analysis of the dilution rate (IC50), its toxicity was reduced from an average of 75% when this set and when it freshly mixed. BC Sealer was not citotoxic. No sealer was able to interfere in the relationship OPG/RANKL. This study showed that the sensitivity of the test used was similar to the cytotoxicity and that the new formulation based bioceramics, BC Sealer was the only group that, in vitro, exhibited no cytotoxicity. The materials did not affect the osteoclastogenesis. |

The materials did not affect the osteoclastogenesis.O objetivo dessa pesquisa foi avaliar, in vitro, a influência de duas novas formulações de cimentos endodônticos: BC Sealer (Brasseler, Estados Unidos da América) e MTA Fillapex (Angelus, Brasil) e do cimento AH Plus (Dentsply, Alemanha) quanto a citotoxicidade em cultura de células humanas e o efeito desses no balanço RANKL/OPG. A partir de fragmentos ósseos descartados pós cirurgias ortopédicas em indivíduos saudáveis, osteoblastos humanos foram cultivados em laboratório e, após confluência, permaneceram expostos ao extratos dos cimentos endurecidos e logo após a manipulação por 24h. Um grupo exposto ao látex e um grupo de células expostas apenas ao meio de cultura serviram como controle positivo e negativo, respectivamente. Para avaliar os efeitos desses cimentos na viabilidade e morfologia de osteoblastos foram empregadas cinco metodologias: teste multiparamétrico para ensaio de citotoxicidade avaliando função mitocondrial (XTT), integridade da membrana (vermelho neutro) e proliferação celular (cristal violeta); teste IC50; microscopia eletrônica de varredura; microscopia de fluorescência para identificar células vitais e citometria de fluxo com a tecnologia do Luminex® utilizando dois painéis humanos que envolvem a osteoclastogênese. Após análises estatísticas, qualitativas e morfológicas, os resultados revelaram que, comparado aos controles, o cimento AH Plus foi citotóxico logo após a manipulação (p<0,05), mas semelhante ao controle negativo quando empregado após o endurecimento. O cimento MTA Fillapex foi citotóxico tanto fresco como após a presa, entretanto, na análise do índice de diluição (IC50), sua toxicidade foi diminuída a partir da média de 75% quando endurecido e logo após a manipulação. O cimento BC Sealer não apresentou toxicidade. Nenhum dos cimentos avaliados promoveu alteração na relação OPG/RANKL em relação ao controle negativo.
