Caracterização física e potencial fisiológico de lotes de sementes de Amburana cearensis (Allemão) A.C. Smith

Resumo: | Amburana cearensis (Allemão) A. C. Smith, belonging to the Fabaceae family, it is a native species of caatinga biome, also known by “amburana” and others same popular names in Brazil according to region, which has potential in the recovery of degraded areas, landscape design and pharmaceutical industry. The present work was developed aimed of physically characterize the seeds, and to study the methodologies for use in tests of emergency, tetrazolium, and conductivity electrical to evaluation of the physiological potential of seeds of A. cearensis deriving from different lots. For the physical characterization, eight repetitions of 100 seeds were weighed and evaluated for length, width and thickness. In germination test the were analyzed the follow variables: percentage, first count, germination speed, length and dry mass of root and shoot. For the emergency test, sowing was carried out in beds, the temperature and relative humidity of 30 °C and 75%, respectively, were analyzed: percentage (first and final count) and emergence speed, length and dry mass of the root and shoot. Moisture was obtained with a triplicate of 5 g at 105 °C for 24 hours in drying oven. The electrical conductivity was conducted with four repetitions of 50 seeds for each lot and the tetrazolium test was accomplished with manually scarified seeds, being immersed directly in distilled water for 24 hours, at 35 oC, and after total removal of the integument. Then, were then immersed in 0.05 % solution of 2,3,5-triphenyltetrazolium chloride salt for three hours at 40 °C under dark conditions. The experimental design was completely randomized with four replications of 25 seeds (50 seeds for electrical conductivity) for each test and lot. For the emergency test, a randomized block design was used. Results indicated great variation in seed length, width and thickness for the different lots used. According to weight of one thousand, it was found a significant variation with emphasis on the seeds represented by the L1 lot with the lowest values compared to the other lots studied. How much the physiological potential, it was found superiority of the seeds represented by lots L1, L4 and L6 for the variables, germination, first count, GSI, seedling length, dry mass and emergence in field. Among the L1 and L6 lots, stood out as more vigorous by the electrical conductivity test. Tetrazolium test showed the difference between each lot vigor levels as well as the type of damage that was done to the seed. |

Smith, pertencente à família Fabaceae, é uma espécie arbórea nativa da Caatinga, encontrada em praticamente toda a América do Sul, conhecida popularmente como: amburana, amburana-de-cheiro, dentre outros nomes populares, a qual tem potencial para uso na recuperação de áreas degradadas, projetos paisagísticos e indústria farmacêutica. O presente trabalho foi desenvolvido com o objetivo de caracterizar fisicamente as sementes, e estudar as metodologias para uso em testes de emergência, tetrazólio e condutividade elétrica, para avaliação do potencial fisiológico de sementes de A. cearensis, provenientes de diferentes lotes. Para a caracterização física oito repetições de 100 sementes foram pesadas e avaliadas quanto ao comprimento, largura e espessura. No teste de germinação, as variáveis analisadas foram: porcentagem, primeira contagem, velocidade de germinação, comprimento e massa seca de raiz e parte aérea. Para o teste de emergência, a semeadura foi realizada em canteiros, a temperatura e umidade relativa do ar de 30 °C e 75%, respectivamente, foram analisadas: porcentagem (final e primeira contagem) e velocidade de emergência, comprimento e massa seca da raiz e parte aérea. O grau de umidade foi obtido com três repetições de 5 g a 105 °C por 24 horas em estufa. A condutividade elétrica foi conduzida com quatro repetições de 50 sementes para cada lote e o teste de tetrazólio foi realizado com sementes escarificadas manualmente, sendo imersas diretamente em água destilada por 24 horas, na temperatura de 35 oC, para posterior retirada total do tegumento. Em seguida, as mesmas foram imersas em solução 2,3,5 cloreto trifenil tetrazólio a 0,05% por três horas, a 40 oC, em condição de escuro. O delineamento experimental foi o inteiramente ao acaso, com quatro repetições de 25 sementes (50 sementes para a condutividade elétrica) para cada teste e lote. Para o teste de emergência, foi utilizado o delineamento em blocos ao acaso. Os resultados indicaram grande variação do comprimento, largura e espessura das sementes referentes aos diferentes lotes utilizados. No tocante ao peso de 1000 sementes, constatou-se uma variação significativa com destaque para as sementes representadas pelo lote L1 com os mais baixos valores comparado aos demais lotes estudados. Quanto ao potencial fisiológico, constatou-se superioridade das sementes representadas pelos lotes L1, L4 e L6 para as variáveis, germinação, primeira contagem, IVG, comprimento de plântulas, massa seca e emergência em campo. Dentre estes lotes, o L1 e L6 se destacaram como mais vigorosos pelo teste de condutividade elétrica.
