Flavonoides rutina e quercetina reduzem a perda óssea alveolar na doença periodontal experimental

Resumo: Periodontitis is an inflammatory disease associated with bone loss and is a frequent cause of tooth loss in adults. Flavonoids are compounds abundant in vegetables and have anti- inflammatory actions. The aim of the present study was to investigate the role of flavonoids rutin and quercetin on the pathogenesis of experimental periodontal disease (EPD). EPD was induced in Wistar rats by placing a nylon thread around the left upper second molar. Flavonoids rutin (25, 50 e 100 mg/kg) and quercetin (75, 150 e 300 mg/kg) were injected intraperitoneally (i.p.) one hour before periodontitis induction and daily for eleven day. The animals were sacrificed on the 11 th day and the parameters analyzed were alveolar bone loss (ABL), histological analysis and leukogram measured before and after challenge (6h and 1, 7 and 11 days). It was observed that the animals with EPD presented a high ABL on the 11 th day. The treatment with rutin or quercetin reduced the ABL. The histological analysis showed a significant reduction of inflammatory cell infiltration and osteoclast number, as well as preservation of the alveolar process and cementum. The animal leukogram with EPD showed leukocytosis with neutrophilia at 6 hours and leukocytosis with elevated mononuclear cells on 7 th and 11 th day. Flavonoids rutin or quercetin reduced the leukocytosis on the 11 th day. These data provide evidence that flavonoids reduce inflammatory process and cementum and bone resorption in experimental periodontitis

Os flavonóides são compostos encontrados em alimentos de origem vegetal que têm mostrado efeitos antiinflamatórios. O objetivo deste tra- balho foi investigar o papel dos flavonóides rutina e quercetina na patogênese da doença periodontal experimental (DPE). DPE foi induzida em ratos Wistar pela inserção de fio de náilon em torno do segundo molar superior esquerdo. Os flavonóides rutina (25, 50 e 100mg/ kg) ou quercetina (75, 150 e 300mg/kg) foram injetados intraperitonealmente 1h antes da indução da DPE e, após esta, diariamente. Os animais foram sacrificados no 11º dia e os seguintes parâmetros foram avaliados: índice de perda óssea (IPO), análise histológica e leucogramas realizados antes e após a cirurgia (6h e 1, 7 e 11 dias). Os animais com DPE apresentaram IPO elevado no 11º dia. O tratamento com rutina ou quercetina diminuiu a perda óssea alveolar. A análise histológica mostrou redução significante do infiltrado celular inflama- tório e do número de osteoclastos, bem como preservação do processo alveolar e cemento. Os leucogramas dos animais com DPE apresentaram leucocitose na 6ª hora às custas de neutrófilos e nos 7º e 11º dias com aumento no número de mononucleares. Os flavonóides rutina ou quercetina reduziram significativamente a leucocitose animais no 11° dia.
