Efeito anti-inflamatório do extrato hidroalcoólico de folhas de Chenopodium ambrosioides L. na bexiga de ratos submetidos à cistotomia

Resumo: | Healing of the bladder has several particularities. The contact of the bladder tissue with urine and its constituents, the need for use of intra-bladder catheters to prevent it from filling and suffer early distension during the healing period means at risk of complications such as infection, fistula and calculus formation. The search for alternative healing products has been constant. In this context, the plant species Chenopodium ambrosioides L., popularly known as mastruz, mastruço and erva-deSanta-Maria, has been reported by the population as antiinflammatory, analgesic and healing, seems a good prospect. The present study investigated the effects of the hydroalcoholic extract (HAE) of leaves of Chenopodium ambrosioides in the healing of the urinary bladder in Wistar rats. Thirty-six rats had been underwent cystotomy and reconstruction of the bladder and then divided into three groups, treated, by gavage, in the first three days postoperatively with: phosphate buffer solution (PBS) (control group); Chenopodium ambrosioides HAE (mastruz group ) in daily dose of 5 mg/kg or oral solution piroxicam (piroxicam group) at a daily dose of 0.3 mg/kg. The animals were sacrificed at 3rd, 7th and 14th days post-surgery, the urinary bladder had been removed and prepared for histopathological and immunohistochemistry analysis to myofibroblast. The rate of adhesions, the production of oxygen peroxide in the peritoneum and nitric oxide in lymph nodes and cellularity of these organs were also surveyed. Chenopodium ambrosioides HAE was able to histologically decrease capillary neoformation and ischemic necrosis, as well as fibroblast proliferation, collagen and chronic inflammation of the bladder when compared to the control group. However, the HAE did not change adhesion, the production of oxygen peroxide and nitric oxide and cellularity of the peritoneum and lymph nodes and myofibroblasts actin productions when compared to the control group. In the present study, Chenopodium ambrosioides HAE showed cronic antiinflamatory and antifibrotic activity on the urinary bladder after cystotomy followed by bladder suture, in rats. |

O contato do tecido vesical com urina e seus constituintes, a necessidade de uso de cateteres intravesicais que a impeça de encher e sofrer distensão precoce durante o período de cicatrização implicam em risco de complicações como infecção, fístula e formação de cálculos. A busca de produtos cicatrizantes alternativos tem sido constante. Neste contexto, a espécie vegetal Chenopodium ambrosioides L., conhecida popularmente como mastruz, mastruço e erva-de-Santa-Maria, que tem sido relatada pela população como anti-inflamatória, analgésica e cicatrizante, parece uma boa perspectiva. O presente estudo investigou os efeitos do extrato hidroalcoólico (EHA) de folhas de Chenopodium ambrosioides na cicatrização da bexiga urinária em ratos da linhagem Wistar. Trinta e seis ratos foram submetidos a cistotomia e cistorrafia, e, em seguida, divididos em três grupos, tratados por gavagem, nos três primeiros dias de pósoperatório com: solução tamponada com fosfato (PBS) (grupo controle); EHA de Chenopodium ambrosioides (grupo mastruz), em dose diária de 5 mg/kg ou solução oral de piroxicam (grupo piroxicam), em dose diária de 0,3 mg/kg. Os animais foram sacrificados no 3 o , 7o e 14o dias pós-cirúrgicos, a bexiga urinária foi retirada e preparada para análise histopatológica e imunohistoquímica para avaliação de miofibroblastos. Foram pesquisados, ainda, o índice de aderências, a produção de peróxido de hidrogênio (H2O2) por células peritoneais e de óxido nítrico (NO) por células dos linfonodos e o número de células no peritônio e linfonodos. O EHA de Chenopodium ambrosioides foi capaz de diminuir a necrose isquêmica e a neoformação capilar, bem como, a proliferação fibroblástica, colagenização e inflamação crônica na parede vesical, quando comparado ao grupo controle. Entretanto, o EHA não alterou o índice de aderências, a produção de peróxido de hidrogênio e óxido nítrico, a celularidade do peritônio e linfonodos e a expressão de alfa-actina em miofibroblastos quando comparado ao grupo controle.
