Polimorfismo do gene da Interleucina 6 (rs1800795) e sua relação com papilomavírus humano em mulheres do nordeste brasileiro

Resumo: Human papillomavirus (HPV) is one of the main risk factors for developing cervical cancer, especially high-risk HPV such as types 16 and 18. In carcinogenesis an inflammatory response occurs at different stages of tumor development. Interleukin 6 (IL6) is a central mediator of inflammation and acts as a pro-inflammatory and anti-inflammatory cytokine in regulating the immune response. The present study aimed to verify the relationship between IL6 polymorphism (rs1800795) and susceptibility to HPV infection and possible cervicovaginal lesions and infections in women in a capital of northeastern Brazil. This is a cross-sectional, analytical, prospective study with 195 sexually active women who used the public health system of São Luís/Maranhão/Brazil. Sociodemographic data, information related to sexual health and pregnancy of the participants were evaluated. The cytopathological analysis was performed using Papanicilaou staining with the Bethesda classification (2014). The detection of IL-6 - 174G>C polymorphism (rs1800795) was performed according to the Real Time Polymerase Chain Reaction (Real Time PCR) technique. To evaluate the relationship between epidemiological and clinical data and the presence of HPV, two groups were used and the results were considered statistically significant when p<0.05. Among the 195 participants, 68 (34.88%) were positive for HPV-DNA and 37 (15.95%) had a diagnosis of cervical cancer. It was found that the distribution of the C allele of IL6 -174G>C (rs1800795) was 2.05%, where 2.05% presented homozygosity (CC) and 26.67% heterozygosity (CG), presenting Hardy-Weinberg equilibrium. There was no relationship between the polymorphism of IL-6 -174G>C (rs1800795) between the groups with and without HPV-DNA. In addition, there was no association with cytopathological changes. However, there was an association between polymorphism and the presence of Trichomonas vaginalis (p<0.05). The data obtained in this study showed no association between the presence of HPV-DNA and the IL6 -174G>C polymorphism (rs1800795) in sexually active Ludovic women, but the polymorphism was associated with the presence of Trichomonas vaginalis.

Na carcinogênese ocorre uma resposta inflamatória em diferentes estágios de desenvolvimento do tumor. A interleucina 6 (IL6) é um mediador central da inflamação e atua como uma citocina pró-inflamatória e anti-inflamatória na regulação da resposta imune. O presente trabalho teve como objetivo verificar a relação entre o polimorfismo da IL6 (rs1800795) e à suscetibilade à infecção por HPV e possíveis lesões e infecções cervico-vaginais em mulheres em uma capital do nordeste brasileiro. Trata-se de um estudo transversal, do tipo analítico, de modo prospectivo com 195 mulheres sexualmente ativas que utilizaram o sistema público de saúde de São Luís/Maranhão/Brasil. Avaliou-se dados sociodemográficos, informações relativas a saúde sexual e a gestação das participantes. A análise citopatológica foi realizada utilizando-se a coloração de Papanicilaou com a classificação Bethesda (2014). A detecção do polimorfismo da IL-6 -174G>C (rs1800795) foi realizado segundo a técnica de reação em cadeia de polimerase por tempo real (PCR Real Time). Para a avaliação da relação entre os dados epidemiológicos e clínicos e a presença de HPV, utilizou-se dois grupos e os resultados considerados estatisticamente significativos quando p<0,05. Dentre as participantes 195, 68 (34,88%) foram positivas para o DNA-HPV e 37 (15,95%) tinham o diagnóstico de câncer de colo de útero. Verificou-se que a distribuição do alelo C do IL6 -174G>C (rs1800795) foi de 2,05%, onde 2,05% apresentaram em homozigose (CC) e 26,67% em heterozigose (GC), apresentando equilíbrio de Hardy-Weinberg. Não houve relação entre o polimorfismo da IL-6 -174G>C (rs1800795) entre os grupos com e sem DNA-HPV. Além disso, não houve associação com as alterações citopatológicas. Porém, houve associação entre o polimorfismo e a presença de Trichomonas vaginalis (p<0,05).
