Resumo: | The genus Bacteroides includes Gram negative rods, obligate anaerobes, considered to be as members of the indigenous microbiota of humans and other animals. They are often recovered from infectious processes, mainly of intra-abdominal origin. Among Bacteroides species, Bacteroides fragilis should be highlighted considering that it is commonly recovered from clinical specimens, expresses several virulence abilities and frequently exhibits antimicrobial resistance. The ability to synthesize bacteriocins is widely spread among bacteria. We aimed to evaluate the expression of antibacterial activity by B. fragilis, including the investigation of the antagonistic activity spectrum and the extraction of a proteinaceous antagonistic substance. Antagonistic expression was evaluated by the double layer method. A total of 19 bacterial isolates, members of the intestinal microbiota or enteropathogens were included as indicator strains. Among the 57 tests conducted nine gave positive results. Isoantagonistic and heteroantagonistic activity were detected. One of the three test strains, B. fragilis D51, was selected for further steps of the investigation. Interfering factors were excluded suggesting the expression of a bacteriocin. The producer strain was submitted to protein extraction and among the intracellular fractions obtained only C30 expressed antagonistic activity (800 AU/mL). C30 lost its activity following treatment with proteinase K confirming its proteinaceous nature. Taken into consideration the relevance of the theme as well as the scarcity of data regarding this subject we believe that this investigation should be continued in order to allow a better understanding about B. fragilis virulence and bacteriocins relevance. |

O grupo é frequentemente recuperado de processos infecciosos, principalmente intra-abdominais. Dentre as diversas espécies incluídas no gênero, destaca-se Bacteroides fragilis, espécie frequentemente recuperada de espécimes clínicos, capaz de expressar uma ampla gama de habilidades de virulência e multirresistência a drogas antimicrobianas. Diversos microrganismos são capazes de expressar substâncias antimicrobianas de natureza proteica, denominadas bacteriocinas, relevantes do ponto de vista ecológico e com grande potencial de aplicação biotecnológico. Este estudo foi realizado com o objetivo de avaliar a atividade antibacteriana de B. fragilis, mais especificamente, pesquisar o espectro de ação antagonista e extrair uma substância proteica antagonista. A expressão de antagonismo foi avaliada pelo método de difusão em sobrecamada, empregando-se, como reveladoras, 19 amostras de bactérias membros da microbiota intestinal indígena ou enteropatogênicas. Dentre os 57 testes realizados, nove apresentaram resultados positivos, sendo observados isoantagonismo e heteroantagonismo. Dentre as três amostras produtoras testadas, selecionou-se aquela com espectro de ação mais amplo, B. fragilis D51, para as etapas subsequentes do estudo. A exclusão de fatores interferentes, quais sejam bacteriófagos, ácidos graxos, clorofórmio e peróxido de hidrogênio, sugeriu a síntese de substância antagonista tipo bacteriocina. A amostra produtora foi submetida a extração proteica por precipitação com concentrações crescentes de sulfato de amônio, obtendo-se as frações intracelulares C30, C50 e C80. Apenas C30 expressou antagonismo; o título de atividade foi 800 UA/mL. O extrato C30 foi inativado pelo tratamento com proteinase K, confirmando sua natureza proteica. Os resultados demonstram a expressão de substância(s) antagonista(s) de natureza proteica por B. fragilis.
