Resumo: | Prednisone is one of the drugs of the glucocorticoid class, which has properties to alter the metabolism of carbohydrates, lipids and proteins, deregulating a series of physiological, biochemical and immunological processes, and is easily obtainable by the population, since it does not require doctor's prescription. It should be noted that there is a shortage of prednisone withdrawal protocols and, as this is a widely used drug, it is necessary to understand the biological interference of these different withdrawal protocols. The aim of this study was to evaluate the biochemical effects resulting from prednisone treatment in different drug reduction protocols in Wistar rats. Four groups of Wistar rats were used, each group with a type of reduction treatment regimen, namely: the Prednisone Group (GP, N=7) which received Prednisone throughout the treatment, Withdrawal Group (GR, N= 7) who received Prednisone 5mg/Kg for 14 days, and from the 15th day the treatment was replaced by SyrSpend® (drug vehicle) until the 28th day. Withdrawal Group A (GRA, N=7) received Prednisone 5mg/Kg for 14 days and underwent dose reduction and then withdrawal, where, on the 15th day there was a dose reduction to 2.5mg/Kg and on the 21st day, replacement by SyrSpend®. And the Withdrawal Group B (GRB, N=7) received Prednisone 5mg/Kg for 14 days and that, like the GRA Group, suffered a dose reduction, however, from the 15th day, there was a 50% reduction in the dose of Prednisone every 3 days and on the 27th day SyrSpend® was replaced. Drug treatment consisted of a single daily administration. Leukogram was performed on days 0, 14 and 28 of treatment and on the last day, the rats were euthanized and then blood was collected for serum assessments of Total cholesterol, triglycerides, high-density lipoproteins (HDL), nonHDL cholesterol (n -HDL), Glucose, albumin, total proteins, aspartate aminotransferase (AST) and alanine aminotransferase (ALT), in addition to total and differential leukocyte counts and Pearson's correlation between total leukocytes and lymphocytes with biochemical biomarkers. Results were considered statistically significant when p<0.05. There were no differences in the values of leukogram counts and serum protein, lipid and glucose biochemicals. However, the GRB group showed an increase in serum ALT activity compared to all other groups; in addition, in all groups with reduction and withdrawal protocol (GR, GRA and GRB) AST activity values increased in relation to continuous treatment. In this way, it was possible to conclude that the treatment of Wistar rats submitted to different treatment regimes shows that there is still no difference between these protocols to find an ideal model of weaning from the drug, opening space for the development of new methodologies, especially in the study higher doses and extended treatment times. |

Ressalta-se que há uma escassez de protocolos de retirada da prednisona e, por este ser um medicamento amplamente utilizado, necessário se faz compreender as interferências biológicas destes diferentes protocolos de retirada. O objetivo deste estudo foi avaliar os efeitos bioquímicos resultantes do tratamento com prednisona em diferentes protocolos de redução do medicamento em ratos de linhagem Wistar. Foram utilizados quatro grupos de ratos Wistar, cada grupo um com um tipo de regime de tratamento de redução, sendo estes: o Grupo Prednisona (GP, N=7) o qual recebeu Prednisona durante todo o tratamento, Grupo Retirada (GR, N=7) que recebeu Prednisona 5mg/Kg por 14 dias, e a partir do 15º dia o tratamento foi substituído por SyrSpend® (veículo do medicamento) até os 28 dias. Grupo Retirada A (GRA, N=7) recebeu Prednisona 5mg/Kg por 14 dias e foi submetido a redução de dose e depois a retirada, onde, ao 15º dia houve redução da dose para 2,5mg/Kg e no 21º dia, substituição por SyrSpend®. E o Grupo Retirada B (GRB, N=7) recebeu Prednisona 5mg/Kg por 14 dias e que assim como o Grupo GRA, sofreu redução da dose, porém, a partir do 15º dia, ocorreu a redução de 50% da dose de Prednisona a cada 3 dias e ao 27º dia ocorreu substituição por SyrSpend®. O tratamento com a droga foi de única administração diária. Foi realizado leucograma nos dias 0, 14 e 28 de tratamento e no último dia, os ratos foram eutanasiados e em seguida houve a coleta de sangue para avaliações séricas de colesterol Total, triglicerídeos, lipoproteínas de alta densidade (HDL), colesterol não-HDL (n-HDL), Glicose, albumina, proteínas totais, aspartato aminotransferase (AST) e alanina aminotransferase (ALT), além da contagem total e diferencial de leucócitos e correlação de Pearson entre leucócitos totais e linfócitos com os biomarcadores bioquímicos. Os resultados foram considerados de significância estatística quando p<0,05. Não houve diferenças dos valores das contagens de leucograma e dos bioquímicos séricos proteicos, lipídicos e de glicose. Entretanto, o grupo GRB apresentou um aumento da atividade sérica da ALT em comparação a todos os outros grupos. Ademais, todos os grupos com protocolo de redução e retirada (GR, GRA e GRB) aumentaram os valores de atividade da AST em relação ao tratamento contínuo.
