Resumo: The creation of the PCR technique was a landmark for science, driving technological growth and opening new possibilities for the diagnosis of human diseases, especially infectious diseases. However, the advent of this new technology restricted its use, almost exclusively, to research laboratories and specialized centers in molecular biology, due to its dependence of expensive and specific devices and the complexity of procedures. As knowledge advanced and new technologies emerged, the limitations of the PCR technique were also targets for improvement. In this way, since 1990s, few years after the PCR technique advent, new approaches were created, allowing the execution of the reaction under isothermal conditions and demanding less or none specific devices. Since 1990s, several isothermal amplification techniques have been created and applied in diagnosis of infectious diseases, such as: NASBA™ (Nucleic Acid Sequence-Based Amplification), 3SR (Self-Sustained Sequence Replication), SDA (Strand Displacement Amplification), RCA (Rolling Circle Amplification), HDA (Helicase-Dependent Amplification), CPA (Cross Priming Amplification), PSR (Polymerase Spiral Reaction) and LAMP (LoopMediated Isothermal Amplification). These techiniques have created a range of alternatives and new concepts of nucleic acids amplification. Here we gave a special highlight for LAMP, a highly sensitive and specific technique whose improvement has created new methods for results visualization and new technologies to make it simpler, efficient and less expensive.

Mas o advento dessa nova tecnologia deixava restrito o seu uso, quase que exclusivamente, aos laboratórios de pesquisa e centros especializados em biologia molecular, devido à sua dependência de aparelhos onerosos e específicos e à complexidade dos procedimentos realizados. Conforme o conhecimento em biologia molecular avançou e novas tecnologias foram surgindo, as limitações atreladas à técnica de PCR também foram alvos de incrementos. Dessa forma, desde a década de 1990, poucos anos após a criação da técnica de PCR, novas abordagens foram criadas, permitindo que a reação fosse realizada em condições isotérmicas e com menor necessidade de aparelhos específicos.Desde a década de 1990, diversas técnicas de amplificação isotérmica foram criadas e aplicadas no contexto do diagnóstico de doenças infecciosas, dentre elas: NASBA™ (Nucleic Acid Sequence-Based Amplification), 3SR (Self-Sustained Sequence Replication), SDA (Strand Displacement Amplification), RCA (Rolling Circle Amplification), HDA (HelicaseDependent Amplification), CPA (Cross Priming Amplification), PSR (Polymerase Spiral Reaction) e LAMP (Loop-Mediated Isothermal Amplification). Estas técnicas representam uma gama de alternativas e novos conceitos de amplificação de ácidos nucleicos.
