Polimorfismos da p53 em mulheres com e sem expressão de RNAm E6/E7 de HPV

Objective: To evaluate the association between the presence of p53 polymorphisms and expression of human papillomavirus (HPV) E6/E7 mRNA. Methods: cross-sectional study that analyzed 175 cervical samples of women 16 to 69 years old who underwent HPV E6/E7 mRNA testing (NucliSENS® EasyQ® HPV). The samples were divided into three groups: positive (n=75), those with expression of HPV E6/E7 mRNA plus positive high-risk HPV hybrid capture (HC); negative (n=52), those without expression of HPV E6/E7 mRNA but positive high-risk HPV HC; and control (n=48), those without expression of HPV E6/E7 mRNA plus negative high-risk HPV HC. The presence of p53 codons 11, 72 and 248 polymorphisms was analyzed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. Results: for p53 codon 72 polymorphisms, the frequency of arginine/arginine homozygosis was significantly higher in positive group (49.3%) than negative (32.7%) and control groups (20.8%, p=0.002*). Also, the frequency of arginine allele was significantly higher in the positive group (67.3%) than negative (53.8%) and control groups (38.5%, p<0.001*). The odds ratio of arginine/arginine homozygote women present expression of HPV E6/E7 mRNA (positive group) compared to other women (negative and control groups) was 2.633 times (95% confidence interval: 1.399-4.954, p=0.003*). The frequency of codon 72 arginine/arginine homozygosis remained significantly higher in positive group compared to negative and control groups in women ?30 years old. There were no statistically significant differences neither in the frequency of homozygote and heterozygote genotypes nor in the allele frequency of p53 codons 11 and 248 polymorphisms among positive, negative, and control groups. Conclusion: the presence of p53 codon 72 arginine/arginine homozygosis was significantly associated to the expression of HPV E6/E7 mRNA.
