Extração de antocianinas dos frutos do açai da Mata Atlântica (Euterpe edulis Martius) e sua atuação nas atividades antioxidante e antiaterogênica em camundongos ApoE -/-

Resumo: | Açaí is a fruit of the palm trees of the genus Euterpe Martius. In the Amazon region the açaí drink is obtained from the pulp of the fruits of açaizeiro (Euterpe oleracea Martius). In the region of the Atlantic Forest can be obtained from the açaí fruit from Juçara (Euterpe edulis Martius). This is a palm characteristic of the Atlantic Forest occurring from the Rio Grande do Sul to Bahia, with main distribution in the Atlantic coast. Currently, E. edulis is a palm species threatened with extinction. There are few reports of beneficial effects and composition of the fruits from this species. It has been argued that positive effect on health is related to the presence of unsaturated fatty acids, minerals and phenolic compounds such as anthocyanins in this fruit. In this study we tested the best solvent for the extraction of anthocyanins and evaluated their antioxidant and antiatherogenic effects by adding the extract of freeze-dried açaí pulp to the diet of ApoE-/- mice divided into four experimental groups. The group C+ (positive control) received the standard AIN-93M diet, the group 2% and 6% received extracts of açaí in the AIN-93M diet at 2% and 6% respectively, and the group SINV received the drug simvastatin on the estimated amount of 50mg/kg/day for mice. A fifth group, C57BL6 mice (negative control) received standard AIN-93M diet for 75 days of experiment. A reduction in the levels of antioxidant enzymes (superoxide dismutase and catalase) in the liver tissue of mice treated with açaí, suggesting that this activity may have been suppressed due to the synergistic effect of anthocyanins. There was no reduction in the evolution of atherosclerotic plaques for the treated groups compared to the positive control, but açaí was able to reduce glucose, total cholesterol and LDL, showing that there was no relationship between the plaques evolution and cholesterol levels during this treatment time. The results showed that the extract of freeze-dried açaí improved antioxidant activity, blood glucose and lipid profile, but was not able to minimize the development of atherosclerotic plaque in ApoE-/- mice during the treatment period. |

Na região amazônica a bebida de açaí é obtida a partir da polpa dos frutos do açaizeiro (Euterpe oleracea Martius). Na região da Mata Atlântica, pode-se obter o açaí a partir dos frutos da Juçara (Euterpe edulis Martius). Esta é uma palmeira endêmica da Mata Atlântica que ocorre desde o Rio Grande do Sul até a Bahia, com distribuição principal na orla Atlântica. Atualmente a E. edulis é uma espécie de palmeira ameaçada de extinção. Existem poucos relatos de composição e efeitos benéficos dos frutos desta espécie.Tem-se discutido o seu efeito positivo na saúde, relacionando-se à presença de ácidos graxos insaturados, minerais e compostos fenólicos como as antocianinas. Neste estudo testou-se o melhor solvente para a extração de antocianinas e avaliaram-se seus efeitos antioxidante e antiaterogênico adicionando o extrato da polpa do açaí liofilizado à dieta de camundongos ApoE-/- divididos em quatro grupos experimentais. O Grupo C+, controle positivo, recebeu a dieta padrão AIN-93M, os grupos 2% e 6%, receberam os extratos de açaí na dieta AIN-93M na proporção de 2% e 6%, respectivamente. O grupo Sinv, recebeu o fármaco Sinvastatina na quantidade estimada de 50mg/kg/dia por camundongo. Um quinto grupo, controle negativo, com camundongos C57Bl6 recebeu dieta padrão AIN-93M, durante 75 dias de experimento. Observou-se redução dos níveis das enzimas antioxidantes (catalase e superóxido dismutase) no tecido hepático dos camundongos tratados com açaí, sugerindo que esta atividade pode ter sido suprimida devido ao efeito sinergista das antocianinas. Não houve redução na evolução das placas de ateroma para os grupos que receberam tratamento quando comparados ao controle positivo, porém o açaí foi capaz de reduzir glicose, colesterol total e LDL, demonstrando que não há relação entre a placa e os níveis de colesterol durante este tempo de tratamento.
