Hidroxiapatita associada à lignina no preenchimento de falha óssea experimental na tíbia de coelhos

Resumo: | The propose of this work was to evaluate synthetic hydroxyapatite with lignin in solid form as substitute in bone regeneration. Twenty rabbits, from New Zealand race, clinically healthy were used. The animals were divided in two groups with 10 animals each one. Group one received hydroxyapatite associated lignin and group two didn t received the implant. After customary anesthetic and surgical protocol in all the animals, a skin incision was made on lateral surface in the right pelvic member in their third middle. A circular bone fragment with 4,5 mm of diameter was retreated from the tibia in all groups, and the bone defect was filled up as a protocol for each treatment group. The animals were submitted to a daily clinical inspection to observe inflammatory reaction and painful sensibility. Radiographs were taken immediately after the surgery and at four, eight, 15, 30 and 60 days after surgery to evaluate bone healing. At same time, two animals per time period were sacrificed and fragments for histopathological evaluations through optical microscopy were collected. The samples were fixed in 10% formol and processed as customary technical in histopathology. The clinical results showed that most of the animals had a normal clinical evolution without infection or rejection of the implants. The results of the X-rays showed a decreasing of opacity on the bone defect until they look like regular bone in the groups that received hidroxyapatite. In the control group, the X-rays revealed increasing of opacity on the bone defect. The histological results showed normal evaluation of the bone regeneration in control group and in the treated one, although it was more precocious in the group that received hidroxyapatite until the 30th day observation. Therefore, the use of hidroxyapatite associated with lignin as bone substitute showed to be efficient, without inflammatory reaction and showing to be biocompatible. |

Para tanto, foram utilizados vinte coelhos, clinicamente sadios, da raça Nova Zelândia. Os animais foram divididos em dois grupos, com 10 animais em cada. O grupo um recebeu hidroxiapatita associada à lignina e o grupo dois não recebeu o implante. Após a realização de protocolo anestésico rotineiro, foi feita uma incisão de pele no terço médio do membro pélvico direito, expondo a tíbia. Um fragmento ósseo circular de 4,5 mm de diâmetro foi retirado, com o auxílio de uma broca trefina, nos animais dos dois grupos, e o defeito foi preenchido de acordo com o protocolo de cada grupo. Os animais foram submetidos à avaliação clinica diária com o objetivo de se observar reação inflamatória e sensibilidade dolorosa. Foram realizadas radiografias imediatamente após o procedimento cirúrgico e aos quatro, oito, 15, 30 e 60 dias do pós-operatório para se avaliar o processo de regeneração óssea. Nas mesmas datas, exceto no dia da cirurgia, dois animais de cada grupo foram sacrificados objetivando a coleta de material para histologia. As amostras foram fixadas em formol 10%, tamponado, e processadas segundo técnicas rotineiras de histologia. Os resultados clínicos mostraram que a maioria dos animais teve evolução clinica normal, sem indícios de infecção ou de rejeição do implante. As radiografias mostraram, no grupo tratado, um decréscimo de radiopacidade do defeito, até que a imagem ficasse semelhante ao osso. Já no grupo controle, observou-se o inverso, ou seja, aumento gradual da radiopacidade do defeito. Os resultados histológicos mostraram evolução normal no processo de regeneração óssea nos dois grupos. Entretanto, nos animais tratados, o processo de reparação óssea se mostrou mais precoce nas observações de 15 e 30 dias, não havendo diferença entre os grupos na observação subseqüente.
