Resumo: | The Phlebolomus and Lutzomyía sandflies are found in the Old and New World, respectively. Their adult females are haematophagous and important vectors of viruses, bacteria and various species of Leíshmanía. Despite their importance, some aspects of their physiology are poorly studied yet, including the organization of your fat body (FB). The FB is a multifunctional organ and its functions include storage of nutrients (such as proteins, carbohydrates and lipids). The aim of this study was to investigate the FB of adult females of Lutzomyía longípaüaís and Phlebolomus papalasí, including the subcellular changes that occur in the FB trophocytes after blood feeding, and also study the effect of polyclonal antisera against the midgut peritrophic matrix proteins (i.e. chitinase and peritrophin) in the lipid accumulation in the trophocytes. Two groups of females were used of each species: 45h after blood feeding and the control, fed with sucrose solution (20-30%). Samples were processed for light and electron microscopy, and for morphometric analysis, trophocyte lipid droplets were considered. For the females that receive the blood diet with antibodies, the lipid droplet areas were measured 24, 48 and 72h after feeding. The FB is quite similar between the two species in terms of location, organization and cellular composition. The FBs are distributed throughout the whole insect body and are organized as parietal and perivisceral lobes. The FBs include the trophocytes and oenocytes. The trophocytes are the most abundant cells, rich in mitochondria and RER mainly in blood fed females. The oenocytes are rich in smooth endoplasmic reticulum, suggesting their role in lipid processing. Although L. longípahyís and P. papatasí are autogenous and anautogenous, respectively, they share subcellular changes observed after blood feeding, even in the former species the blood acquisition is not mandatory for the egg development. In addition, in non-blood fed and in blood fed individuals, there is no significant differences in the lipid droplet area in the trophocytes of L. longípahyís and P. papatasí. The treatment with antibodies has been shown to influence the supply of nutrients by FB. In L. longípaüaís 24h after feeding showed no significant difference. Conversely, 48h after blood feeding significant differences in the lipid droplet control is almost two times higher than the antiperitrophin and antichitinase fed females However, among the females treated with these two antibodies there is no difference in terms of lipid droplet area. Seventy two h after feeding, the droplets area in antisera fed females was almost two times higher than the control. In P. papatasí, the lipid droplets area 24h after the treatment with antiserum against chitinase is almost three-fold greater compared to the control, while 48h and 72h after feeding the area was not significantly different among females treated with antisera against the chitinase and peritrophin. Our results suggest that the antibodies were able to negatively interfere with the digestion process in L. longípalpís and P. papatasí from 48h followed the blood meal, even if they were developed from P. papatasz'. This work constitutes and provides important information for the better understanding of the sandflies FB biology. |

Suas fêmeas são hematófagas e importantes vetores de vírus, bactérias e várias espécies de Leíshmanía. Apesar de sua importância, alguns aspectos da morfofisiologia desses insetos permanecem pouco estudados, incluindo a organização do seu corpo gorduroso (CG). O CG é um órgão multifuncional, cujas funções incluem o armazenamento de nutrientes (proteínas, carboidratos e lipídeos). O objetivo deste trabalho foi descrever o CG de fêmeas adultas de Lutzomyía longípahyís e Phlebotomus papatasí, estudar as alterações subcelulares que ocorrem quando submetidas a diferentes dietas e estudar o efeito da administração de anticorpos antiquitinase e antiperitrofina na dieta sanguínea no acúmulo de lipídeos nos trofócitos. Dois grupos de fêmeas foram usados para as duas espécies: 45h após o repasto sanguíneo e alimentadas com solução de sacarose (20-30%), estudados através das microscopias óptica e eletrônicas de varredura e transmissão e pelas análises de morfometria, onde foram medidas as áreas das gotículas lipídicas dos trofócitos. A abordagem morfométrica também foi empregada em fêmeas submetidas às dietas sanguíneas contendo dois anticorpos contra a matriz peritrófica: antiquitinase e antiperitrofina. O CG é bastante semelhante entre as duas espécies em termos de localização, organização e composição celular, sendo subdivididos em parietal (logo abaixo do tegumento) e perivisceral. As células que compõem o CG são os trofócitos e os enócitos, sendo os primeiros muito mais abundantes, ricos em mitocôndrias e RER nos insetos alimentados com sangue. Os enócitos são células ricas em REL, reforçando a hipótese delas estarem envolvidas na síntese de lipídeos. Apesar de serem espécimes anautógenas e autógenas (L. longípahyís e P. papatasí, respectivamente), elas compartilham alterações subcelulares observadas mediante a dieta sanguínea, mesmo que no primeiro caso ela não seja essencial para o desenvolvimento dos ovócitos. Não houve diferenças significativas no tamanho da área das gotículas de lipídeos entre fêmeas tratadas com os dois anticorpos e as do grupo controle. Por outro lado, 48h após o repasto a área das gotículas de lipídeo do controle é quase duas Vezes maior do que os espécimes tratados com antiperitrofina e antiquitinase, porém, entre as fêmeas tratadas com os anticorpos não houve diferença. Em 72h após o repasto, a área das gotículas de fêmeas tratadas com antiperitrofina foi quase duas Vezes maior do que do controle e dos tratados com antiquitinase. Em P. papatasí, a área das gotículas 24h após o repasto tratadas com antiquitinase é quase o triplo em relação ao controle e em tomo de duas Vezes maior que às tratadas com antiperitrof1na. Em 48h e 72h após o repasto houve diferença significativa apenas entre as fêmeas tratadas com antiquitinase e antiperitrof1na. Nossos resultados sugerem que os anticorpos foram capazes de interferir indiretamente no armazenamento de lipídeos em L. longípahaís e P. papalasí a partir das 48h seguidas do repasto sanguíneo.
