Resumo: The aim of this study was to investigate the association between polymorphisms in the IL-1a and IL-1b genes and severity of chronic periodontitis in Brazilians. 113 non-smoking subjects over 25 years (mean age 41.6) were divided according to the severity level of periodontal disease: 44 healthy individuals (control group), 31 subjects with moderate and 38 with severe periodontitis. DNA was obtained from epithelial cells through a mouthwash with 3 % glucose and scraping of oral mucosa. The samples were analyzed for IL-1a (- 889) and IL-1b (+3953) polymorphisms using PCR-RFLP. The significance of the differences in the frequencies of the polymorphisms in the control and groups with periodontitis was assessed by Chisquare test (p<0.05). No significant differences in the allele and genotype frequencies were found among the groups. It was concluded that neither polymorphisms IL-1a (-889) and IL-1b (+3953) nor the composite genotype were associated with the severity of chronic periodontal disease in the study population.
