Oxidative stress and neuroinflammation in neurodegenerative diseases: possible neuroprotective effect of agathisflavone

Resumo: | Oxidative stress and neuroinflammation are gaining prominence as essential contributing factors in neurodegenerative diseases. In this context, the flavonoids have powerful anti-inflammatory and antioxidant properties that can serve as therapy in these diseases. A representative of this group is the biflavonoid, agathisflavone, which in several studies has demonstrated biological actions that deserve attention for the pharmaceutical industry in the therapy of neurodegenerative diseases. Through a systematic review of the literature, we analyzed the scientific/experimental publications published between 2011 and 2020 in the databases of PubMed and ScienceDirect, with the aim of systematizing the important findings on agathisflavone that may contribute to its therapeutic approach. Data were extracted from 11 studies that documented important therapeutic actions that can help in the treatment of neurodegenerative diseases, such as anti-inflammatory activity, immunomodulator, neurogenesis induction, free radical neutralization, neuroprotection and neuronal death reduction. From the data obtained, it was observed that agathisflavone has promising characteristics to act as an adjuvant neuroprotector for the treatment of neurodegenerative diseases; however, it is necessary more in-depth and specific studies, so that its therapeutic value is evidenced. |

En este contexto, los flavonoides tienen poderosas propiedades antiinflamatorias y antioxidantes que pueden servir como terapia en estas enfermedades. Un representante de este grupo es el biflavonoide, la agatisflavona, que en varios estudios ha demostrado acciones biológicas que merecen atención por parte de la industria farmacéutica en la terapia de las enfermedades neurodegenerativas. Mediante un examen sistemático de la bibliografía, se analizaron las publicaciones científicas/experimentales publicadas entre 2011 y 2020 en las bases de datos de PubMed y ScienceDirect, a fin de sistematizar los importantes hallazgos sobre la agatisflavona que pueden contribuir a su enfoque terapéutico. Se extrajeron datos de 11 estudios que documentaron importantes acciones terapéuticas que pueden ayudar en el tratamiento de enfermedades neurodegenerativas, como: actividad antiinflamatoria, inmunomoduladora, inducción de neurogénesis, neutralización de radicales libres, neuroprotección y reducción de la muerte neuronal. A partir de los datos obtenidos, se observó que la agatisflavona tiene características prometedoras para actuar como neuroprotector adyuvante en el tratamiento de las enfermedades neurodegenerativas; sin embargo, es necesario realizar estudios más profundos y específicos para que se pueda demostrar su valor terapéutico.O estresse oxidativo e a neuroinflamação veem ganhando destaque como fatores contribuintes essenciais nas doenças neurodegenerativas. Nesse contexto os flavonoides possuem potentes propriedades anti-inflamatórias e antioxidantes que podem servir de terapia nestas doenças. Um representante deste grupo é o biflavonóide, agatisflavona, que em diversos estudos vem demonstrando ações biológicas que merecem atenção para a indústria farmacêutica na terapia das doenças neurodegenerativas. Através de uma revisão sistemática da literatura, analisou-se as publicações cientificas/experimentais publicadas entre 2011 e 2020, nas bases de dados da PubMed e ScienceDirect, com o objetivo de sistematizar os achados importantes sobre a agatisflavona que possam contribuir para sua abordagem terapêutica. Foram extraídos dados de 11 estudos que documentaram ações terapêuticas importantes que podem auxiliar no tratamento de doenças neurodegenerativas, como: atividade anti-inflamatória, imunomoduladora, indução de neurogênese, neutralização de radicais livres, neuroproteção e redução da morte neuronal.
