Evaluation of the antimicrobial potential of the Ethanolic extract of Cecropia pachystachya t. (Embaúba) Leaves

Resumo: The present study aims to evaluate the antimicrobial potential of the ethanol extract of Cecropia pachystachya (embaúba) leaves. To achieve the microbiological tests used were Gram-positive bacteria S. aureus ATCC 29213, S. aureus ATCC 25925, S. pyogenes 1033B and the clinical isolate from the Mycology Laboratory of the Federal University of Pernambuco, C. albicans 7098. The extract was prepared using the newly collected material (5:10; p/v) in ethanolic solution with concentrations of 50 and 70%, remaining in contact with the solvent for 72 hours. After the period, the eluent was filtered, stored and kept in a refrigerator at 4°C until use. The evaluation of antibacterial activity was carried out in triplicate by agar diffusion technique and to perform the tests, the strains were recovered on Nutritive Agar (NA) and incubated in an oven at 35 ± 2 ºC for 24h. For the analysis of the inhibitory activity of the extracts, the diameters of the inhibition halos (mm) were read. The extract from Embaúba demonstrated activity against the tested bacterial strains, whose results obtained were halos that ranged from 12mm to 18mm. However, more in vitro and in vivo studies are needed to evaluate the compounds present in the extract in isolation in order to elucidate the antimicrobial activity, considering the possibility of the development of drugs with spectrum of action against Gram-positive bacteria and yeasts.

Para la realización de las pruebas microbiológicas se utilizaron las bacterias Gram positivas S. aureus ATCC 29213, S. aureus ATCC 25925, S. pyogenes 1033B, y el aislado clínico del Laboratorio de Micología de la Universidad Federal de Pernambuco, C. albicans 7098. El extracto se preparó utilizando el material recién recolectado (5:10; p/v) en una solución alcohólica con concentraciones de 50 y 70%, permaneciendo en contacto con el solvente por 72h. Después del período, el eluyente se filtró, se almacenó y se mantuvo en un refrigerador a 4° C hasta su uso. La evaluación de la actividad antibacteriana se realizó por triplicado mediante la técnica de difusión en agar y para la realización de las pruebas, las cepas se recuperaron en agar nutriente y se incubaron en estufa a 35 ± 2 ºC durante 24h. Para el análisis de la actividad inhibidora de los extractos se leyeron los diámetros de los halos de inhibición (mm). El extracto de Embaúba demostró actividad frente a las cepas bacterianas ensayadas, cuyos resultados obtenidos fueron halos que variaron de 12mm a 18mm. Sin embargo, se necesitan más estudios in vitro e in vivo para evaluar los compuestos en el extracto de forma aislada con el fin de dilucidar la actividad antimicrobiana, considerando la posibilidad de desarrollar fármacos con espectro de acción frente a bacterias Gram positivas y levaduras.O presente estudo tem como objetivo avaliar o potencial antimicrobiano do extrato etanólico de folhas da Cecropia pachystachya(embaúba). Para a realização dos ensaios microbiológicos foram utilizadas as bactérias Gram-positivas S. aureus ATCC 29213, S. aureus ATCC 25925, S. pyogenes 1033B, e o isolado clínico C. albicans 7098 oriundos do Laboratório de Micologia da Universidade Federal de Pernambuco. O extrato foi preparado utilizando o material recém coletado (5:10; p/v) em solução alcoólica com concentrações de 50 e 70%, permanecendo em contato com o solvente por 72h. Após o período, o eluente foi filtrado, armazenado e mantido em refrigerador a 4°C até sua utilização. A avaliação da atividade antibacteriana foi realizada em triplicata através da técnica de difusão em ágar e para realização dos testes, as cepas foram recuperadas em ágar nutritivo e incubadas em estufa a 35 ± 2 ºC por 24h. Para análise da atividade inibitória dos extratos, foi realizada a leitura dos diâmetros dos halos de inibição (mm). O extrato da Embaúba demonstrou atividade contra as linhagens bacterianas testadas, cujos resultados obtidos foram halos que variaram de 12mm a 18mm.
