ISSR markers are effective to access genetic diversity in germplasm of wheat (Triticum aestivum)

Resumo: | Know the genetic variability between wheat cultivars (Triticum aestivum) is of great importance for breeding programs. Thus, this work aimed to use ISSR markers to access genetic diversity between contrasting wheat accessions. For this, 32 accessions of wheat and one access of triticale from the Active Bank of Wheat Germplasm of the Institute of Rural Development of Paraná (IDR-Paraná) were evaluated. A total of 66 polymorphic loci were amplified from 10 ISSR primers with an average loci variation per primer of 6.6. Genetic similarity ranged from 0.29 to 0.93, with an average of 0.57. The resulting dendrogram from the data was divided into four groups, in which it was observed that the genotypes LD 111202 and LD 111203 showed a high level of genetic similarity among themselves. The Bayesian cluster analysis formed four groups, partially in agreement with the UPGMA hierarchical cluster. ISSR markers were able to discriminate morphologically distinct and similar accessions and provide consistent information on the genetic variability between wheat genotypes that will help in breeding programs. |

Por lo tanto, este trabajo tuvo como objetivo utilizar marcadores ISSR para acceder a la diversidad genética entre accesiones de trigo contrastantes. Para eso, se evaluaron 32 accesiones de trigo y un acceso de triticale del Banco Activo de Germoplasma de Trigo del Instituto de Desarrollo Rural de Paraná (IDR-Paraná). Se amplificó un total de 66 loci polimórficos a partir de 10 cebadores ISSR con una variación promedio de 6,6 loci por cebador. La similitud genética osciló entre 0,29 y 0,93, con un promedio de 0,57. El dendrograma resultante de los datos se dividió en 4 grupos, en los que se observó que los genotipos LD 111202 y LD 111203 mostraron una alta tasa de similitud genética entre sí. El análisis de conglomerados bayesianos formó cuatro grupos, parcialmente de acuerdo con el conglomerado jerárquico de UPGMA. Los marcadores ISSR pudieron discriminar accesiones distintas y similares, además de proporcionar información consistente sobre la variabilidad genética entre genotipos de trigo que ayudará en los programas de mejoramiento.Conhecer a variabilidade genética existente entre cultivares de trigo (Triticum aestivum) é de grande importância para os programas de melhoramento. Desta forma, esse trabalho teve como objetivo utilizar marcadores ISSR para acessar a diversidade genética entre acessos contrastantes de trigo. Para isso, foram avaliados 32 acessos de trigo e um acesso de triticale provenientes do provenientes do Banco Ativo de Germoplasma de Trigo do Instituto de Desenvolvimento Rural do Paraná (IDR-Paraná). A partir de 10 primers de ISSR foram amplificados um total de 66 loci polimórficos com variação média de loci por primer de 6,6. A similaridade genética variou de 0,29 a 0,93, com média de 0,57. O dendrograma resultante mostrou 4 grupos, no qual observou-se que os genótipos LD 111202 e LD 111203 apresentaram alto índice de similaridade genética entre si. A análise bayesiana de agrupamento formou quatro grupos, parcialmente concordantes com o agrupamento hierárquico de UPGMA.
