Study of the effect of Ginkgo biloba and quercetin extracts on the mouse vas deferens

Resumo: Ginkgo biloba extract (GBE) has antioxidant, anti-inflammatory, and neuroprotective actions, and has a relaxing effect on smooth muscle organs. The vas deferens (DD) conducts the spermatozoa during ejaculation by the contraction of its muscles, and changes in this contraction can lead to disorders such as premature ejaculation (PE). PE is ejaculation that always or almost always occurs before or about 1 minute after vaginal penetration. Thus, our aim was to study the effect of GBE under the contraction of the DD of mice. The effect of GBE, here called GB2, or quercetin, was tested against contraction with potassium chloride. The effect of these substances was tested in the presence of nifedipine. We demonstrate that GB2, quercetin and nifedipine are able to inhibit the phasic and tonic components of the contraction. The effect of GB2 on the tonic component was enhanced in the presence of quercetin. Comparing the tonic component between GB2, Nifedipine+GB2 and Quercetin+GB2 groups, we observed that in the presence of nifedipine or quercetin, the effect of GB2 was potentiated. In the comparison of the Quercetin and Nifedipine+Quercetin groups, nifedipine potentiated the effect of quercetin under both components. These results indicate that GB2 and quercetin are able to inhibit some mechanism involved with the generation of DD contraction, and that this effect is not solely dependent on voltage-dependent calcium channel inhibition. These findings point out that GBE and quercetin, may be pharmacological strategies for the treatment of PE.

El conducto deferente (DD) conduce los espermatozoides durante la eyaculación mediante la contracción de sus músculos, y las alteraciones de esta contracción pueden provocar trastornos como la eyaculación precoz (EP). La EP es una eyaculación que siempre o casi siempre se produce antes o aproximadamente un minuto después de la penetración vaginal. Por lo tanto, nuestro objetivo era estudiar el efecto de la GBE en la contracción del DD de los ratones. El efecto del GBE, aquí denominado GB2, o quercetina, se probó contra la contracción con cloruro de potasio. El efecto de estas sustancias se probó en presencia de nifedipina. Demostramos que el GB2, la quercetina y el nifedipino son capaces de inhibir los componentes fásicos y tónicos de la contracción. El efecto del GB2 sobre el componente tónico se potenció en presencia de quercetina. Al comparar el componente tónico entre los grupos GB2, Nifedipino+GB2 y Quercetina+GB2, observamos que, en presencia de nifedipino o quercetina, el efecto del GB2 se potenciaba. En la comparación de los grupos de quercetina y nifedipino+quercetina, el nifedipino potenció el efecto de la quercetina en ambos componentes. Estos resultados indican que el GB2 y la quercetina son capaces de inhibir algún mecanismo implicado en la generación de la contracción DD, y que este efecto no depende únicamente de la inhibición del canal de calcio dependiente de voltaje. O efeito do GBE, aqui denominado GB2, ou quercetina, foi testado frente a contração com cloreto de potássio. O efeito destas substâncias foi testado na presença da nifedipina. Demonstramos que GB2, quercetina e nifedipina são capazes de inibir os componentes fásico e tônico da contração. O efeito de GB2, sob o componente tônico, foi potencializado, na presença da quercetina. Comparando o componente tônico entre os grupos GB2, Nifedipina+GB2 e Quercetina+GB2, observamos que, na presença da nifedipina ou da quercetina, o efeito de GB2 foi potencializado. Na comparação dos grupos Quercetina e Nifedipina+Quercetina, a nifedipina potencializou o efeito da quercetina sob ambos os componentes. Esses resultados indicam que GB2 e a quercetina são capazes de inibir algum mecanismo envolvido com a geração da contração do DD, e que esse efeito não é dependente exclusivamente da inibição dos canais de cálcio voltagem dependente.
