Toxicity and genotoxicity of drugs used in the Covid-19 pandemic through the Allium cepa bioassay

Resumo: During the Covid-19 pandemic, there was an increase in the commercial consumption of the medicines Hydroxychloroquine and Ivermectin for the treatment of the disease due to the positive results found in in vitro tests. However, there is no scientific evidence that these drugs, in safe dosages, are effective for the pathology in humans. In this sense, this study aimed to evaluate the toxicity and genotoxicity of drugs through the Allium cepa bioassay. Thus, for the positive control, 800mg of Paracetamol was used and for the negative control, treated water. The drugs Hydroxychloroquine and Ivermectin showed toxic potential at their usual dosages. In addition, one of the bulbs treated with Ivermectin showed morphological changes in its roots. The antimalarial drug (Hydroxychloroquine) did not present a genotoxic risk at the dosages tested, but it poses a risk to human health when administered in excess, without medical advice, or in association with other drugs, such as azithromycin. The drug Ivermectin presented a genotoxic risk in two of the four concentrations tested, showing an increase in the number of micronuclei according to the increase in the dose. Therefore, for the consumption of these drugs, responsible medical monitoring is necessary, in addition to following the appropriate recommendations. To date, the correct vaccination protocol is the only effective method of prophylaxis.

Sin embargo, no existe evidencia científica de que estos fármacos, en dosis seguras, sean efectivos para la patología en humanos. En ese sentido, este estudio tuvo como objetivo evaluar la toxicidad y genotoxicidad de fármacos a través del bioensayo Allium cepa. Así, para el control positivo se utilizaron 800 mg de Paracetamol y para el control negativo agua tratada. Los fármacos Hidroxicloroquina e Ivermectina mostraron potencial tóxico en sus dosis habituales. Además, uno de los bulbos tratados con Ivermectina presentó cambios morfológicos en sus raíces. El fármaco antipalúdico (Hidroxicloroquina) no presentó riesgo genotóxico a las dosis probadas, pero presenta riesgo para la salud humana cuando se administra en exceso, sin consejo médico, o en asociación con otros fármacos, como la azitromicina. El fármaco Ivermectina presentó riesgo genotóxico en dos de las cuatro concentraciones probadas, mostrando un aumento en el número de micronúcleos conforme aumentaba la dosis. Por tanto, para el consumo de estos fármacos es necesario un seguimiento médico responsable, además de seguir las recomendaciones oportunas. Hasta la fecha, el protocolo de vacunación correcto es el único método eficaz de profilaxis.Durante a pandemia da Covid-19, houve um aumento no consumo comercial dos medicamentos Hidroxicloroquina e Ivermectina para o tratamento da doença devido aos resultados positivos encontrados em testes in vitro. Entretanto, não há indícios científicos de que estes medicamentos, em dosagens seguras, sejam eficazes para a patologia em seres humanos. Neste sentido, este trabalho teve como objetivo avaliar a toxicidade e genotoxicidade dos medicamentos através do bioensaio Allium cepa. Com isso, para controle positivo, foi utilizado 800mg de Paracetamol e para o negativo, água filtrada. Os fármacos Hidroxicloroquina e Ivermectina apresentaram potencial tóxico em suas dosagens usuais. Além disso, um dos bulbos tratados com Ivermectina apresentou alterações morfológicas em suas raízes. O medicamento antimalárico (Hidroxicloroquina) não apresentou risco genotóxico nas dosagens testadas, mas oferece risco à saúde humana quando administrada em excesso, e sem recomendação médica, ou com associação com outros medicamentos, tal como a azitromicina. O fármaco Ivermectina apresentou risco genotóxico em duas das quatro concentrações testadas, apresentando aumento do número de micronúcleos de acordo com o aumento da dose. Portanto, para consumo desses medicamentos, é necessário um acompanhamento médico responsável, além de seguir as recomendações adequadas.
